IgG in vitro ELISA (Enzyme-Linked Immunosorbent Assay) kit is designed for the quantitative measurement of IgG protein in bovine cell culture supernatant, milk, plasma and serum.
In this assay the IgG present in samples reacts with the anti-IgG antibodies which have been adsorbed to the surface of polystyrene microtitre wells. After the removal of unbound proteins by washing, anti-IgG antibodies conjugated with horseradish peroxidase (HRP), are added. These enzyme-labeled antibodies form complexes with the previously bound IgG. Following another washing step, the enzyme bound to the immunosorbent is assayed by the addition of a chromogenic substrate, 3,3',5,5'-tetramethylbenzidine (TMB). The quantity of bound enzyme varies directly with the concentration of IgG in the sample tested; thus, the absorbance, at 450 nm, is a measure of the concentration of IgG in the test sample. The quantity of IgG in the test sample can be interpolated from the standard curve constructed from the standards, and corrected for sample dilution.
Bovine IgG also known as bovine immunoglobulin G is a prominent antibody found in bovine species that plays a central role in their immune response. It has a molecular mass of approximately 150 kDa. Bovine IgG is expressed significantly in the blood and colostrum of cows. It is an important part of the immune system helping identify and neutralize pathogens such as bacteria and viruses. The different subtypes of bovine IgG like IgG1 and IgG2 possess specific roles in recognizing a variety of antigens.
The functions of bovine immunoglobulin G involve critical immunological processes. It interacts with Fc receptors on immune cells including macrophages and neutrophils to trigger phagocytosis and other immune responses. Bovine antibodies enable these cells to effectively attack and remove pathogens. Although it is not part of a large multimeric complex it synergizes with other components of the immune system such as complement proteins to enhance the clearance of antigens.
Bovine IgG plays an integral role in both humoral immune response and complement pathways. Within the humoral immune response bovine immunoglobulin interacts with proteins such as complement proteins C1q and C3 to mediate complement activation facilitating opsonization and pathogen lysis. This antibody supports immune system communication and coordination directing immune cells to areas where pathogens are present. These processes ensure a coordinated response to pathogens and help maintain immune homeostasis.
Bovine immunoglobulin G is a critical player in preventing infectious diseases like bovine respiratory disease complex and mastitis. A deficiency or dysfunction in bovine IgG can lead to increased susceptibility to these infections. Moreover the antibody's lack of function or presence in the context of these diseases can correlate with other proteins such as cytokines which are involved in inflammatory responses. Understanding and manipulating bovine IgG levels can offer therapeutic strategies for mitigating these diseases thereby improving cattle health and productivity.
