Bovine IgG ELISA Kit ab205078 is a sandwich ELISA to measure Bovine IgG in serum, plasma, cell culture supernatant, milk with a sensitivity of 1.245 ng/mL.
How the assay works:
In this assay the IgG present in samples reacts with the anti-IgG antibodies which have been adsorbed to the surface of polystyrene microtitre wells. After the removal of unbound proteins by washing, anti-IgG antibodies conjugated with horseradish peroxidase (HRP), are added. These enzyme-labeled antibodies form complexes with the previously bound IgG. Following another washing step, the enzyme bound to the immunosorbent is assayed by the addition of a chromogenic substrate, 3,3',5,5'-tetramethylbenzidine (TMB). The quantity of bound enzyme varies directly with the concentration of IgG in the sample tested; thus, the absorbance, at 450 nm, is a measure of the concentration of IgG in the test sample. The quantity of IgG in the test sample can be interpolated from the standard curve constructed from the standards, and corrected for sample dilution.

1. Add standard or sample solution to each well. Incubate at room temperature.
2. Aspirate and wash each well. Add prepared HRP labeled secondary detector antibody. Incubate at room temperature.
3. Aspirate and wash each well. Add Chromogen Substrate Solution to each well.
4. Measure immediately.

Biological function summary
The functions of bovine immunoglobulin G involve critical immunological processes. It interacts with Fc receptors on immune cells including macrophages and neutrophils to trigger phagocytosis and other immune responses. Bovine antibodies enable these cells to effectively attack and remove pathogens. Although it is not part of a large multimeric complex it synergizes with other components of the immune system such as complement proteins to enhance the clearance of antigens.

Bovine IgG plays an integral role in both humoral immune response and complement pathways. Within the humoral immune response bovine immunoglobulin interacts with proteins such as complement proteins C1q and C3 to mediate complement activation facilitating opsonization and pathogen lysis.
