cAMP SimpleStep ELISA® kit is a single-wash 90 min competitive ELISA designed for the quantitative measurement of cAMP in Cell and Tissue extracts. Quantitate cAMP with 0.22 ng/ml sensitivity.
SimpleStep ELISA® technology employs capture conjugates conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to competitive ELISA allows the formation of the antibody-analyte complex in a single step, significantly reducing assay time. Free cAMP was spiked into the following samples and diluted in a 2-fold dilution series in 1X Cell Extraction Buffer PTR. Undiluted samples are 0.5 mg/ HepG2 cell extract, Jurkat cell extract and Human kidney extract. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2).

Comparison of native cAMP concentration in treated versus control cell extract. HepG2 cells were treated with 30 µM Forskolin and 1 mM IBMX diluted in cell culture media for 15 minutes, scraped from the plate and then extracted with 1X Cell Extraction Buffer PTR. Mock control HepG2 cells accompanied cell treatment. Concentrations of cAMP were measured in duplicate and interpolated against the standard curve. Results show the mean cAMP concentration in control cell extract was 0.69 ng/mL compared to 11.20 ng/mL in the forskolin- and IBMX-treated sample.
Interpolated concentration of cAMP was measured in duplicate at different sample concentrations. Undiluted samples are 0.25 mg/mL HepG2 Cell Extract. Cells were treated for 15 minutes with 30 µM Forskolin and 1 mM IBMX. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). Sample dilutions are made in 1X Cell Extraction Buffer PTR.
