Sandwich ELISA - Canine Clusterin ELISA Kit (AB325457)

Interpolated concentration of native Clusterin was measured in duplicate at different sample concentrations in 96-well vs. 384-well plates. Undiluted samples are 1 : 5,000 canine serum. The interpolated dilution factor corrected values (to neat sample) are plotted (mean +/- SD, n=2). Sample dilutions are made in Sample Diluent NS.

Sandwich ELISA - Canine Clusterin ELISA Kit (AB325457)

Interpolated concentration of native Clusterin was measured in duplicate at different sample concentrations. Undiluted samples are as follows : canine serum 1 : 2,500, canine plasma (citrate) 1 : 5,000, canine plasma (EDTA) 1 : 5,000, and canine plasma (heparin) 1 : 5,000. The interpolated dilution factor corrected values (to neat sample) are plotted (mean +/- SD, n=2). Sample dilutions are made in Sample Diluent NS.

Product details

Canine Clusterin ELISA Kit (ab325457) is a rapid single-wash sandwich 90-min ELISA to measure Canine Clusterin in Serum, Plasma – Citrate, Plasma – EDTA, Plasma – Heparin, Cell Culture Supernatant. This SimpleStep sensitivity is 0.38 ng/ml.

How the assay works

Canine Clusterin ELISA Kit (ab325457) SimpleStep ELISA® employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details.

Assay Specificity

Our SimpleStep ELISA® kits use recombinant monoclonal antibodies rigorously validated to ensure the highest level of consistency and reproducibility, improved sensitivity and specificity and ease of scalability and security of supply.
Please refer to our protocol booklet for more details.

Canine Clusterin (ab325457) protocol summary

1. Mix : add samples / standards to the wells together with the capture and detector antibody cocktail. Incubate 1 hr at room temperature
2. Wash
3. Add TMB development solution - incubate for 10 min
4. Add Stop solution
5. Read the results on a plate reader at 450 nm

Save your precious samplesOur Canine Clusterin ELISA Kit only requires 12.5µL sample.

Target data

Functions as extracellular chaperone that prevents aggregation of non native proteins. Prevents stress-induced aggregation of blood plasma proteins. Inhibits formation of amyloid fibrils by APP, APOC2, B2M, CALCA, CSN3, SNCA and aggregation-prone LYZ variants (in vitro). Does not require ATP. Maintains partially unfolded proteins in a state appropriate for subsequent refolding by other chaperones, such as HSPA8/HSC70. Does not refold proteins by itself (By similarity). Binding to cell surface receptors triggers internalization of the chaperone-client complex and subsequent lysosomal or proteasomal degradation (PubMed : 11697889). When secreted, protects cells against apoptosis and against cytolysis by complement : inhibits assembly of the complement membrane attack complex (MAC) by preventing polymerization of C9 pore component of the MAC complex. Intracellular forms interact with ubiquitin and SCF (SKP1-CUL1-F-box protein) E3 ubiquitin-protein ligase complexes and promote the ubiquitination and subsequent proteasomal degradation of target proteins. Promotes proteasomal degradation of COMMD1 and IKBKB. Modulates NF-kappa-B transcriptional activity (By similarity). Following stress, promotes apoptosis (By similarity). Inhibits apoptosis when associated with the mitochondrial membrane by interference with BAX-dependent release of cytochrome c into the cytoplasm. Plays a role in the regulation of cell proliferation. An intracellular form suppresses stress-induced apoptosis by stabilizing mitochondrial membrane integrity through interaction with HSPA5. Secreted form does not affect caspase or BAX-mediated intrinsic apoptosis and TNF-induced NF-kappa-B-activity (By similarity). Secreted form act as an important modulator during neuronal differentiation through interaction with STMN3 (By similarity). Plays a role in the clearance of immune complexes that arise during cell injury (By similarity).
