Product details

Canine HGF ELISA kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Canine HGF in serum, plasma and cell culture supernatants.

This assay employs an antibody specific for Canine HGF coated on a 96-well plate. Standards and samples are pipetted into the wells and HGF present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Canine HGF antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of HGF bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

The hepatocyte growth factor (HGF) also known as scatter factor (SF) is a protein with a mass of approximately 83 kDa. This protein is mostly expressed in the liver but it can also be found in other organs such as the kidney and the lungs. HGF acts mechanically by binding to the c-Met receptor activating intracellular signaling mechanisms necessary for various physiological processes. Researchers often use HGF ELISA kits to measure its levels in biological samples and study its various roles.
Biological function summary

HGF plays an essential role in cellular growth regeneration and healing processes across different tissues. It is not part of a complex but functions as a single-chain polypeptide that undergoes cleavage into its active form. HGF facilitates cell proliferation and migration; this is especially important during organ regeneration and repair processes. Receptor tyrosine kinase that transduces signals from the extracellular matrix into the cytoplasm by binding to hepatocyte growth factor/HGF ligand. Regulates many physiological processes including proliferation, scattering, morphogenesis and survival. Ligand binding at the cell surface induces autophosphorylation of MET on its intracellular domain that provides docking sites for downstream signaling molecules. Following activation by ligand, interacts with the PI3-kinase subunit PIK3R1, PLCG1, SRC, GRB2, STAT3 or the adapter GAB1. Recruitment of these downstream effectors by MET leads to the activation of several signaling cascades including the RAS-ERK, PI3 kinase-AKT, or PLCgamma-PKC. The RAS-ERK activation is associated with the morphogenetic effects while PI3K/AKT coordinates prosurvival effects. During embryonic development, MET signaling plays a role in gastrulation, development and migration of muscles and neuronal precursors, angiogenesis and kidney formation. In adults, participates in wound healing as well as organ regeneration and tissue remodeling. Also promotes differentiation and proliferation of hematopoietic cells (By similarity)..
