Type II interferon produced by immune cells such as T-cells and NK cells that plays crucial roles in antimicrobial, antiviral, and antitumor responses by activating effector immune cells and enhancing antigen presentation. Primarily signals through the JAK-STAT pathway after interaction with its receptor IFNGR1 to affect gene regulation. Upon IFNG binding, IFNGR1 intracellular domain opens out to allow association of downstream signaling components JAK2, JAK1 and STAT1, leading to STAT1 activation, nuclear translocation and transcription of IFNG-regulated genes. Many of the induced genes are transcription factors such as IRF1 that are able to further drive regulation of a next wave of transcription. Plays a role in class I antigen presentation pathway by inducing a replacement of catalytic proteasome subunits with immunoproteasome subunits. In turn, increases the quantity, quality, and repertoire of peptides for class I MHC loading. Increases the efficiency of peptide generation also by inducing the expression of activator PA28 that associates with the proteasome and alters its proteolytic cleavage preference. Up-regulates as well MHC II complexes on the cell surface by promoting expression of several key molecules such as cathepsins B/CTSB, H/CTSH, and L/CTSL (By similarity). Participates in the regulation of hematopoietic stem cells during development and under homeostatic conditions by affecting their development, quiescence, and differentiation (By similarity).

Abcam's IFN gamma canine ELISA Kit (ab193684) is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of canine IFN gamma in serum, plasma (collect plasma using EDTA and Citrate as an anticoagulant. Heparin is not recommended for use in this assay) and cell culture supernatants.
This assay employs an antibody specific for canine IFN gamma coated on a 96-well plate. Standards and samples are pipetted into the wells and the immobilized antibody captures IFN gamma present in the samples. The wells are washed and biotinylated anti-canine IFN gamma antibody is added. After washing away any unbound biotinylated antibody, an HRP-conjugated streptavidin is pipetted to the wells. After incubation, the wells are again washed, followed by the addition of a TMB substrate solution to the wells. Color will develop in proportion to the amount of IFN gamma bound in each well. Addition of the Stop Solution will change the color from blue to yellow, and the intensity of the color is measured at 450 nm.
Interferon-gamma (IFNG) also known as type II interferon is a cytokine recognized for its role in immune response. It has a molecular mass of approximately 20 kDa. Immune cells mainly T cells and natural killer cells express IFNG. IFNG binds to its receptor IFNGR1 and IFNGR2 initiating a signaling cascade that activates various genes. This cytokine plays a pivotal role in modulating the immune response and enhancing antigen presentation.
Interferon-gamma is an important regulator of innate and adaptive immunity. IFNG possesses the ability to modulate macrophage activity increase antigen presentation and promote differentiation of T helper 1 cells. It drives activation of other immune cells and sustains an adequate immune response. This cytokine does not function as part of a larger complex but interacts with various immune components to amplify the body's defense response.
Interferon-gamma integrates into the JAK-STAT signaling pathway and the IFN-gamma pathway. In the JAK-STAT pathway IFNG encourages phosphorylation of STAT1 which translocates to the nucleus to influence gene expression. It closely associates with proteins like STAT1 and JAK1 during these interactions. IFNG's role in the IFN-gamma pathway involves orchestrating responses against pathogens by inducing specific genes responsible for immune cell activation and pathogen eradication.
Researchers associate interferon-gamma with tuberculosis and autoimmune disorders like multiple sclerosis. In the context of tuberculosis insufficient IFNG response can result in an ineffective immune defense. IL-12 another cytokine interacts with IFNG playing a central role in developing resistance against infection. In autoimmune disorders such as multiple sclerosis IFNG can exacerbate inflammation due to increased immune cell activity potentially leading to tissue damage. Therefore IFNG remains a focal point in understanding and treating these conditions.
