Product details

This cGMP ELISA kit (ab285321, E4717) is based on Competitive-ELISA principle. The micro ELISA plate provided in this kit has been pre-coated with cGMP. During the reaction, cGMP in the sample or standard competes with a fixed amount of cGMP on the solid phase supporter for sites on the Biotinylated Detection Ab specific to cGMP. Excess conjugate and unbound sample or standard are washed from the plate, and Avidin conjugated to Horseradish Peroxidase (HRP) are added to each microplate well and incubated. Then a TMB substrate solution is added to each well.

The enzyme-substrate reaction is terminated by the addition of stop solution and the color change is measured spectrophotometrically at a wavelength of 450 nm.

The concentration of cGMP in the samples is then determined by comparing the OD of the samples to the standard curve.

Cyclic guanosine monophosphate commonly known as cGMP is a second messenger important in various physiological processes. It has a molecular mass of 345.2 g/mol. cGMP is produced as a result of the conversion of GTP by the enzyme guanylate cyclase and is degraded by phosphodiesterases. cGMP is expressed in several tissues including the brain heart and vascular smooth muscle cells. It plays significant roles in cell signaling and cellular communication.
Biological function summary

Cyclic guanosine monophosphate influences vasodilation and neurotransmission. It functions within cellular signaling complexes and often partners with proteins such as protein kinases to achieve its biological effects. cGMP facilitates the relaxation of smooth muscle tissues impacting cardiovascular health by regulating blood vessel dilation. Its role in vascular regulation links it to hypertension where decreased cGMP levels can elevate blood pressure. Additionally in erectile dysfunction cGMP imbalance disrupts proper blood flow regulation necessary for erection. Phosphodiesterase inhibitors such as sildenafil target cGMP pathways to restore normal function in these conditions highlighting the protein’s involvement in disease treatments.
