Product details

COVID-19 S-Protein (S1RBD) Human IgM ELISA Kit (ab276186) is an in vitro indirect ELISA for the quantitative measurement of human IgM antibody against SARSCoV-2 S1 RBD protein in human serum or dried blood.

Standard 96-well plates (12 strips with 8 wells/strip) are coated with the SARS-CoV-2 S1 RBD protein, which combines with the corresponding antibody present in a sample and Positive Control, which used as calibration curve for interpretation purposes. The wells are washed, and biotinylated anti-human IgM antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells, and color develops in proportion to the amount of COVID19 S1 RBD protein human IgM antibody bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

The Positive Control is from an inactivated serum sample or a dried blood sample which contains SARS-COV-2 S1 RBD protein human IgM antibody. We do not know the exact amount of SARS-COV-2 S1 RBD protein human IgM antibody in the Positive Control sample. The Positive Control can be used as a calibration curve for interpretation purposes in different assays.

The COVID-19 Spike Protein often referred to as SARS-CoV-2 Spike Protein is an important structural protein of the virus with a molecular mass around 180 kDa. This protein facilitates viral entry into host cells by binding to the angiotensin-converting enzyme 2 (ACE2) receptor which is primarily expressed in the respiratory tract. It's a trimeric protein composed of S1 and S2 subunits. The S1 subunit contains the receptor binding domain while the S2 subunit mediates membrane fusion.

Immunoglobulins, also known as antibodies, are membrane-bound or secreted glycoproteins produced by B lymphocytes. In the recognition phase of humoral immunity, the membrane-bound immunoglobulins serve as receptors which, upon binding of a specific antigen, trigger the clonal expansion and differentiation of B lymphocytes into immunoglobulins-secreting plasma cells. Secreted immunoglobulins mediate the effector phase of humoral immunity, which results in the elimination of bound antigens (PubMed : 20176268, PubMed : 22158414). The antigen binding site is formed by the variable domain of one heavy chain, together with that of its associated light chain. Thus, each immunoglobulin has two antigen binding sites with remarkable affinity for a particular antigen. The variable domains are assembled by a process called V-(D)-J rearrangement and can then be subjected to somatic hypermutations which, after exposure to antigen and selection, allow affinity maturation for a particular antigen (PubMed : 17576170, PubMed : 20176268). IgM antibodies play an important role in primary defense mechanisms. They have been shown to be involved in early recognition of external invaders like bacteria and viruses, cellular waste and modified self, as well as in recognition and elimination of precancerous and cancerous lesions. The membrane-bound form is found in the majority of normal B cells alongside with IgD. Membrane-bound IgM induces the phosphorylation of CD79A and CD79B by the Src family of protein tyrosine kinases. It may cause death of cells by apoptosis. It is also found in soluble form, which represents about 30% of the total serum immunoglobulins where it is found almost exclusively as a homopentamer. After the antigen binds to the B cell receptor, the secreted form is secreted in large amounts (, PubMed : 16895553).
