Product details

Abcam's DHEA sulfate (DHEA-S) in vitro competitive ELISA (Enzyme-Linked Immunosorbent Assay) kit is designed for the accurate quantitative measurement of DHEA sulfate in plasma and serum.

A 96-well plate has been precoated with anti-DHEA sulphate antibodies. Samples and the DHEA sulfate-HRP conjugate are added to the wells, where DHEA sulfate in the sample competes with the added DHEA sulfate-HRP for antibody binding. After incubation, the wells are washed to remove unbound material and TMB substrate is then added which is catalyzed by HRP to produce blue coloration. The reaction is terminated by addition of Stop Solution which stops the color development and produces a color change from blue to yellow. The intensity of signal is inversely proportional to the amount of DHEA sulfate in the sample and the intensity is measured at 450 nm.

Supplementary information

Dehydroepiandrosterone sulfate (DHEA-S) also known as DHEA sulphate is a sulfated steroid hormone with a mass of approximately 368.5 Da. DHEA-S is predominantly expressed in the adrenal glands though small amounts can be found in the gonads and brain. It is derived from dehydroepiandrosterone (DHEA) through the action of sulfotransferase enzymes. DHEA-S can be measured through various methods including DHEA sulfate ELISA to assess its concentration in biological samples providing insights into adrenal function and related conditions.
Biological function summary

DHEA-S plays an important role in the production of androgens and estrogens which are significant sex hormones. It acts as a reservoir that can be transformed into these active hormones as needed and this conversion occurs in peripheral tissues. DHEA-S does not operate as part of a stable complex but relies on enzymatic conversion in pathways to exert its effects. Its levels naturally decline with age suggesting its involvement in aging processes and age-related functions making it an interesting target for research on health and longevity.

Pathways

DHEA-S is involved in the steroidogenesis pathway where it interconnects with other steroid hormones like testosterone and estradiol. This pathway is essential for the synthesis and regulation of various steroid hormones. DHEA-S is also connected with the adrenal steroid synthesis pathway and it shares metabolic relationships with enzymes such as 3β-hydroxysteroid dehydrogenase and aromatase which aid in the interconversion of steroid precursors to active hormones.

DHEA-S links to several conditions including adrenal insufficiency and polycystic ovary syndrome (PCOS). In adrenal insufficiency it serves as a marker for reduced adrenal function where lower levels indicate impaired hormone synthesis. In PCOS alterations in DHEA-S levels are common as it can lead to excessive androgen production contributing to symptom development. DHEA-S and its levels can also interact with insulin resistance pathways where the interplay with proteins involved in glucose metabolism becomes significant.
