Recombinant Exendin-4 was spiked into the following species samples and diluted in a 2-fold dilution series in Sample Diluent NS in 96-well vs. 384-well plates. Undiluted samples are 25% cell culture media (RPMI containing 10% FBS). The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2).

How the assay works
Exendin-4 SimpleStep ELISA®employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Biological function summary
Exendin-4 enhances insulin secretion helping to regulate blood glucose levels effectively. It mimics the activity of GLP-1 which is involved in stimulating insulin release. Exendin-4 also slows gastric emptying and promotes satiety contributing to reduced food intake. Although the peptide does not form part of a larger complex its interactions directly affect multiple cellular processes relevant to metabolism.
Pathways
Exendin-4 participates in the insulin signaling and glucose homeostasis pathways. It influences insulin receptor substrate proteins thereby playing an important role in maintaining normal blood sugar levels. Exendin-4 modulates the activity of gluconeogenic enzymes connecting it to metabolic pathways involving energy balance and storage.
