Product details

GST His-tag SimpleStep ELISA® kit is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of GST His-tag protein. Quantitate GST His-tag with 3.476 pg/ml sensitivity.

SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:

-Single-wash protocol reduces assay time to 90 minutes or less
-High sensitivity, specificity and reproducibility from superior antibodies
-Fully validated in biological samples
-96-wells plate breakable into 12 x 8 wells strips

A 384-well SimpleStep ELISA® microplate (ab203359) is available to use as an alternative to the 96-well microplate provided with SimpleStep ELISA® kits.

The biological target 'GST Histidine' refers to a protein tag used in protein purification techniques. It is a fusion tag that includes both glutathione S-transferase (GST) and a histidine tag (His-tag). GST a 26 kDa protein facilitates the purification process when attached to proteins of interest aiding in their solubility and stability. It occurs in various organisms notably expressed in E. coli for laboratory purposes. Meanwhile the His-tag comprises a short sequence of histidine residues that allow for metal affinity chromatography. These dual tags come together to create a versatile tool for studying protein functions.
Biological function summary

The GST and His-tags play essential roles in protein purification and detection processes. They don't operate as part of a cellular protein complex but support the isolation of proteins from complex mixtures. The GST part usually aids in protein folding and enhances solubility. The His-tag facilitates binding to nickel or cobalt ions in a column setup allowing for precise purification. His-tag ELISA assays enable scientists to detect tagged proteins with high specificity and quantify their presence in samples.

Pathways

GST in the context of the tag fusion does not participate in biochemical pathways as it usually would in a cellular environment. However in traditional cell biology GST relates to detoxification pathways and the conjugation of reduced glutathione to toxic substrates. Proteins within these pathways like glutathione peroxidase interact with natural GST. In its role as a tag the His component primarily serves to simplify the purification pathway rather than engaging in typical cellular processes.

The GST tag itself does not have direct connections to specific conditions but helps in research and study of various diseases. For proteins tagged with GST-His researchers can examine interactions with proteins involved in disorders like cancer or neurodegenerative diseases. Studies in these areas often include examining interactions between tagged proteins and cellular proteins such as p53 in cancer studies to provide insight into disease mechanisms.
