Sandwich ELISA - GST His-tag ELISA Kit (AB302758)
Recombinant GST His-tag was spiked into the following samples and diluted in a 2-fold dilution series in 1X Cell Extraction Buffer PTR. Undiluted samples are as follows : serum 10%, lauryl maltoside buffer 50%, urea buffer 10%, 10F RPMI 50%, and guanidine buffer 0.1%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2).

GST His-tag SimpleStep ELISA® kit is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of GST His-tag protein. Quantitate GST His-tag with 3.476 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:

Biological function summary
The GST and His-tags play essential roles in protein purification and detection processes. They don't operate as part of a cellular protein complex but support the isolation of proteins from complex mixtures. The GST part usually aids in protein folding and enhances solubility. The His-tag facilitates binding to nickel or cobalt ions in a column setup allowing for precise purification. His-tag ELISA assays enable scientists to detect tagged proteins with high specificity and quantify their presence in samples.
Pathways
GST in the context of the tag fusion does not participate in biochemical pathways as it usually would in a cellular environment. However in traditional cell biology GST relates to detoxification pathways and the conjugation of reduced glutathione to toxic substrates. Proteins within these pathways like glutathione peroxidase interact with natural GST. In its role as a tag the His component primarily serves to simplify the purification pathway rather than engaging in typical cellular processes.
