Helicobacter pylori IgG plus ELISA Kit ab178645 is a sandwich ELISA to measure Helicobacter pylori IgG plus in serum, citrate plasma, heparin plasma with a sensitivity of 1.39 IU/mL.

How the assay works:

The quantitative immunoenzymatic determination of specific antibodies is based on the ELISA (Enzyme-linked Immunosorbent Assay) technique. Microplates are coated with specific antigens to bind corresponding antibodies of the sample. After washing the wells to remove all unbound sample material a horseradish peroxidase (HRP) labelled conjugate is added. This conjugate binds to the captured antibodies. In a second washing step unbound conjugate is removed. The immune complex formed by the bound conjugate is visualized by adding Tetramethylbenzidine (TMB) substrate which gives a blue reaction product. The intensity of this product is proportional to the amount of specific antibodies in the sample. Sulphuric acid is added to stop the reaction. This produces a yellow endpoint colour. Absorbance at 450/620 nm is read using an ELISA microwell plate reader.

Helicobacter pylori IgG plus ELISA Kit ab178645 protocol summary

1. Add 100 µL control, standard or sample to appropriate wells. Incubate for 1 hour at 37°
2. Aspirate and wash each well three times with 300 µL 1X Washing Solution
3. Add 100 µL of HRP conjugate to each well. Incubate for 30 minutes at room temperature. Repeat the washing steps.
4. Add 100 µL TMB Substrate Solution to each well and incubate for 30 minutes at room temperature.
5. Add 100 µL Stop Solution and read OD at 450/620 nm within 30 minutes after addition of the stop solution.

Helicobacter pylori IgG often referred to as H. pylori IgG is an immunoglobulin produced in response to infection by the bacterium Helicobacter pylori. This bacterium is known for its spiral shape and Gram-negative characteristics. The molecular weight of the IgG antibody varies but generally is around 150 kDa. It is primarily expressed in response to colonization in the gastric mucosa which leads to increased immune response detectable in the bloodstream. The detection of H. pylori IgG in serological assays helps in diagnosing active or previous H. pylori infections.
