Helicobacter pylori IgG plus ELISA Kit ab178645 is a sandwich ELISA to measure Helicobacter pylori IgG plus in serum, citrate plasma, heparin plasma with a sensitivity of 1.39 IU/mL.
How the assay works:
The quantitative immunoenzymatic determination of specific antibodies is based on the ELISA (Enzyme-linked Immunosorbent Assay) technique. Microplates are coated with specific antigens to bind corresponding antibodies of the sample. After washing the wells to remove all unbound sample material a horseradish peroxidase (HRP) labelled conjugate is added. This conjugate binds to the captured antibodies. In a second washing step unbound conjugate is removed. The immune complex formed by the bound conjugate is visualized by adding Tetramethylbenzidine (TMB) substrate which gives a blue reaction product. The intensity of this product is proportional to the amount of specific antibodies in the sample. Sulphuric acid is added to stop the reaction. This produces a yellow endpoint colour. Absorbance at 450/620 nm is read using an ELISA microwell plate reader.

Helicobacter pylori IgG plus ELISA Kit ab178645 protocol summary
1. Add 100 µL control, standard or sample to appropriate wells. Incubate for 1 hour at 37°
2. Aspirate and wash each well three times with 300 µL 1X Washing Solution
3. Add 100 µL of HRP conjugate to each well. Incubate for 30 minutes at room temperature. Repeat the washing steps.
4. Add 100 µL TMB Substrate Solution to each well and incubate for 30 minutes at room temperature.
5. Add 100 µL Stop Solution and read OD at 450/620 nm within 30 minutes after addition of the stop solution.

Biological function summary
Antibodies such as IgG are part of the immune response working to neutralize pathogens like Helicobacter pylori. H. pylori IgG is not part of a larger protein complex but it interacts directly with antigens on the surface of the bacterial cells. By binding to these antigens H. pylori IgG plays a critical role in opsonization which facilitates the recognition and destruction of the bacteria by immune cells thereby contributing to host defense mechanisms.

Together these components coordinate to mount an effective immune response against H. pylori as well as other pathogens. The H. pylori infection triggers a cascade in the immune system that leads to the activation of B cells which produce specific IgG antibodies tailored to recognize the pathogen.
