His-tag SimpleStep ELISA® kit is a single-wash 90 min competitive ELISA designed for the quantitative measurement of His-tag in Serum, Cit Plasma, EDTA Plasma, Hep Plasma, Urine, Cell culture supernatant, Cell Extract, and Tissue Extract. Quantitate His-tag with 36.9 ng/ml sensitivity.SimpleStep ELISA® technology employs capture conjugates conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to competitive ELISA allows the formation of the antibody-analyte complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:
-Single-wash protocol reduces assay time to 90 minutes or less
-High sensitivity, specificity and reproducibility from superior antibodies
-Fully validated in biological samples
-96-wells plate breakable into 12 x 8 wells strips
A 384-well SimpleStep ELISA® microplate (Pre-coated 384 well Microplate SimpleStep ELISA® ab203359) is available to use as an alternative to the 96-well microplate provided with SimpleStep ELISA® kits.

Example of His-tag standard curve. OD (450 nm) values (mean +/- SD) are graphed.
Interpolated concentration of native His-tag was measured in duplicate at different sample concentrations. Undiluted sample is E. coli cell extract expressing a protein with multiple tags at 7.5 µg/mL. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). Sample dilutions are made in 1X Cell Extraction Buffer PTR + Enhancer.
His-tag protein was spiked into the following samples and diluted in a 2-fold dilution series in 1X Cell Extraction PTR + Enhancer. Undiluted samples are 4.5% Human serum, plasma (citrate), plasma (EDTA), plasma (heparin), and urine. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2).
His-tag protein was spiked into the following samples and diluted in a 2-fold dilution series in 1X Cell Extraction PTR + Enhancer. Undiluted samples are 4.5% Mouse serum, plasma (citrate), plasma (EDTA), plasma (heparin), and urine. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2).
His-tag protein was spiked into the following samples and diluted in a 2-fold dilution series in 1X Cell Extraction PTR + Enhancer. Undiluted samples are 4.5% HEK293T cell supernatant, and 4.5% Jurkat cell supernatant. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2).
His-tag protein was spiked into the following samples and diluted in a 2-fold dilution series in 1X Cell Extraction PTR + Enhancer. Undiluted samples are 10 µg/mL HEK293T cell extract, 20 µg/mL CHO cell extract, 36 µg/mL Mouse brain extract, 110 µg/mL Mouse heart extract, and 90 µg/mL Mouse liver extract. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2).
His-tag protein was spiked into the following samples and diluted in a 2-fold dilution series in 1X Cell Extraction PTR + Enhancer. Undiluted samples are 4.5% LB Broth media, 2.25% yeast extraction buffer, 0.7% Lauryl Maltoside buffer, 115 mM urea Buffer, and 22.4 mM imidazole elution buffer. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2).
