Human 4-1BBL ELISA Kit is a single-wash 90-min SimpleStep Sandwich ELISA used to quantify Human 4-1BBL with a sensitivity of 2.2 pg/mL. The assay uses a simple mix-wash-read protocol with just one incubation and one wash step.

Interpolated concentrations of native 4-1BBL in human cell culture supernatant samples.

The concentrations of 4-1BBL were measured in duplicates, interpolated from the 4-1BBL standard curves and corrected for sample dilution. Undiluted samples are as follows : Raji (PMA treated) cell culture supernatant 50% and Raji (untreated) cell culture supernatant 100%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean 4-1BBL concentration was determined to be 143.7 pg/mL in Raji (PMA treated) cell culture supernatant, and 59.2 pg/mL in Raji (untreated) cell culture supernatant.

Human 4-1BBL ELISA Kit ab246536 is a rapid single-wash 90-min ELISA kit to measure Human 4-1 BBL in Citrate plasma, Cell culture supernatant, Serum. This SimpleStep ELISA® sensitivity is 5.64 pg/mL.

How the assay works

Human 4-1 BBL SimpleStep ELISA® employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Assay Specificity

This kit is designed for the quantification of Human 4-1BBL. The standard protein in this kit is the extracellular domain of Human 4-1BBL. Native signal was detected in cell culture supernatant. Spiked protein experiments were used to validate serum and plasma (citrate) sample types. Neat pooled serum and plasma (citrate) samples from healthy donors was measured in duplicate. All values were below the detectable range of the assay. Saliva, urine, milk, CSF, cell extract, and tissue extract samples have not been tested with this kit. This kit is incompatible with plasma (EDTA) and plasma (heparin) samples.

Human 4-1BBL ELISA Kit ab246536 protocol summary:

1. Mix: add samples/standards to the wells together with the capture and detector antibody cocktail. Incubate for 1 hr at room temperature
2. Wash
3. Add TMB development solution - incubate for 10 min
4. Add stop solution
5. Read the results on a plate reader at 450nm
