Serum of ten individual healthy human male donors was measured in duplicate. Interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean ADAM10 concentration was determined to be 2.06 ng/mL with a range of 0.25 – 8.20 ng/mL.

Interpolated concentration of native ADAM10 was measured in duplicate at different sample concentrations. Undiluted samples are stimulated Jurkat supernatant 100%. Jurkat cells were stimulated for 36 hours with 1 µg/mL LPS and 100 ng/mL IFN gamma. The interpolated dilution factor corrected values (to neat sample) are plotted (mean +/- SD, n=2). Sample dilutions are made in Sample Diluent NS.

How the assay works

Human ADAM10 ELISA Kit, Chemiluminescent (ab326004) SimpleStep Ignite™ ELISA employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step during a 30 min only incubation, significantly reducing assay time. By utilizing a chemiluminscent detection method this improves the sensitivity and dynamic range when compared to traditional colorimetric ELISAs.

Cleaves the membrane-bound precursor of TNF at '76-Ala-|-Val-77' to its mature soluble form.
