Sandwich ELISA - Human AKR1B10 ELISA Kit (AB254506)
Interpolated concentrations of native AKR1B10 in human A549 cell extract based on a 0.31 μg/mL extract load.
The concentrations of AKR1B10 were measured in duplicate and interpolated from the AKR1B10 standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2).

Product details
Human AKR1B10 ELISA Kit (ab254506) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of AKR1B10 protein in cell lysate. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human AKR1B10 with 10.3 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:
- Single-wash protocol reduces assay time to 90 minutes or less
- High sensitivity, specificity and reproducibility from superior antibodies
- Fully validated in biological samples
- 96-wells plate breakable into 12 x 8 wells strips
A 384-well SimpleStep ELISA® microplate (ab203359) is available to use as an alternative to the 96-well microplate provided with SimpleStep ELISA® kits.
Aldo-Keto Reductase Family-1 member B10 (AKR1B10), also referred to as Aldose Reductase-like protein, is an 316- amino acid enzyme encoded by the AKR1B10 gene. AKR1B10 belongs to the aldo/keto reductase superfamily, where AKR1B10’s function is to catalyze the NADPH-dependent reduction of carbonyls to the corresponding alcohols. Found to be highly expressed in the small intestine and colon, AKR1B10 is thought to be responsible in detoxifying dietary and lipid derived carbonyls and may play a role in liver carcinogenesis. AKR1B10 has no counterpart in mouse or rat species.

Biological function summary
AKR1B10 protects cells from disorders caused by toxic aldehydes. It is important in lipid biosynthesis through the reduction of retinaldehyde to retinol which is part of the retinoic acid metabolism. AKR1B10 forms part of the aldo-keto reductase superfamily consisting of multiple related enzymes that regulate various cellular processes. This enzyme class is involved in stabilizing the oxidoreduction balance within cells.
Pathways
AKR1B10 engages in lipid metabolism and the protection against oxidative stress. It is an important player in the retinoid metabolism and signaling pathway where it collaborates with enzymes like retinaldehyde dehydrogenase. Additionally AKR1B10 is involved in the polyol pathway although its exact role here continues to be studied. The enzyme's relationship with NADPH suggests it could intersect with systems managing cellular responses to oxidative stress.
