Sandwich ELISA - Human AKT1 ELISA Kit (AB214023)
Interpolated concentrations of native AKT1 in human MCF7, HEK 293T, and HeLa cell extract samples based on 75 μg/mL, 500 μg/mL, and 250 μg/mL extract loads, respectively.
The concentrations of AKT1 were measured in duplicate and interpolated from the AKT1 standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean AKT1 concentration was determined to be 6,370.4 pg/mL, 10,792.3 pg/mL, and 10,157.2 pg/mL in MCF7, HEK 293T, and HeLa cell extract samples, respectively.

Human AKT1 ELISA Kit (ab214023) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of AKT1 protein in cell culture extracts. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human AKT1 with 27.3 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:

AKT1 is one of 3 closely related serine/threonine-protein kinases (AKT1, AKT2 and AKT3) called the AKT kinases. These kinases regulate many processes including metabolism, proliferation, cell survival, growth and angiogenesis. The AKT kinases mediate these processes through serine and/or threonine phosphorylation of a range of downstream substrates. Over 100 substrate candidates have been reported so far, but for most of them, no isoform specificity has been reported.
Each AKT family member contains an N-terminal pleckstrin homology (PH) domain, a central kinase domain, and a C-terminal regulatory domain. Mouse and rat AKT1 exhibit 98% amino acid identity to human AKT1.

Biological function summary
AKT1 regulates a broad spectrum of cellular functions through signaling pathways. It is an important player in the phosphatidylinositol 3-kinase (PI3K) and AKT pathway forming part of a complex involved in promoting cell survival and growth. AKT1 interacts closely with other proteins such as mTOR influencing cellular metabolism and autophagy. The phosphorylation state of AKT1 is critical for its activity with modifications like p-AKT S473 impacting its interaction with cellular substrates.
Pathways
AKT1 participates in the PI3K/AKT/mTOR signaling cascade instrumental in cell proliferation and metabolism. AKT1 integrates signals from insulin and growth factors modulating pathways that control cell growth and glucose uptake. This pathway involves proteins like mTOR and phospho-AKT which coordinate cellular responses to external stimuli. AKT1’s phosphorylation at serine 473 and the involvement of AKT1 E17K a gain-of-function mutation influence these pathways significantly.
