Sandwich ELISA - Human alpha 1 Acid Glycoprotein ELISA Kit (AB243675)
Interpolated concentrations of native alpha 1 Acid Glycoprotein in human urine, milk, saliva and HepG2 cell culture supernatant samples.
The concentrations of alpha 1 Acid Glycoprotein were measured in duplicates, interpolated from the alpha 1 Acid Glycoprotein standard curves and corrected for sample dilution. Undiluted samples are as follows : urine 1 : 800, milk 1 : 500, saliva 1 : 800 and HepG2 cell culture supernatant 3.5%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean alpha 1 Acid Glycoprotein concentration was determined to be 6.85 μg/mL in neat urine, 5.34 μg/mL in neat milk, 8.42 μg/mL in neat saliva and 0.23 μg/mL in neat HepG2 cell culture supernatant.

Sandwich ELISA - Human alpha 1 Acid Glycoprotein ELISA Kit (AB243675)
Interpolated concentrations of native alpha 1 Acid Glycoprotein in human serum and plasma samples.
The concentrations of alpha 1 Acid Glycoprotein were measured in duplicates, interpolated from the alpha 1 Acid Glycoprotein standard curves and corrected for sample dilution. Undiluted samples are as follows : serum 1 : 80,000, plasma (citrate) 40,000, plasma (EDTA) 1 : 80,000 and plasma (heparin) 1 : 80,000. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean alpha 1 Acid Glycoprotein concentration was determined to be 821.2 μg/mL in neat serum, 479.5 μg/mL in neat plasma (citrate), 881.7 μg/mL in neat plasma (EDTA) and 767.8 μg/mL in neat plasma (heparin).

Product details
Human alpha 1 Acid Glycoprotein ELISA Kit ab243675 is a rapid single-wash 90-min Sandwich ELISA to measure Human alpha 1 Acid Glycoprotein in cell culture supernatant, citrate plasma, EDTA plasma, heparin plasma, milk, saliva, serum, urine. This SimpleStep sensitivity is 68.5 pg/mL.
How the assay works
Human alpha 1 Acid Glycoprotein SimpleStep ELISA®employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details.
Assay Specificity
Our SimpleStep ELISA® kits use recombinant monoclonal antibodies rigorously validated to ensure the highest level of consistency and reproducibility, improved sensitivity and specificity and ease of scalability and security of supply.
Please refer to our protocol booklet for more details.

We offer the antibody pair used in this kit in a BSA and Azide-free format, ready for conjugation:
- Anti-Alpha 1 Acid Glycoprotein/AGP antibody [EPR21185-101] - BSA and Azide free (Capture) ab259512

Alpha 1 Acid Glycoprotein is a secreted glycoprotein that functions as transport protein in the blood stream. It binds various ligands in the interior of its beta-barrel domain. It also binds synthetic drugs and influences their distribution and availability in the body. Alpha 1 Acid Glycoprotein appears to function in modulating the activity of the immune system during the acute-phase reaction.

Biological function summary
AGP plays a role in modulating the immune response and maintaining homeostasis. As an acute-phase protein its levels rise significantly during inflammation infection and trauma where it acts to modulate the immune system. AGP does not exist as part of a larger protein complex but interacts with plasma lipoproteins and other glycoproteins aiding in its function of maintaining vascular integrity during acute-phase reactions.
Pathways
Alpha 1 Glycoprotein serves as an important component in the inflammatory response and drug-binding pathways. In the inflammatory pathway AGP interacts with cytokines and acute-phase reactants adjusting the body's inflammatory processes. It is also involved in the binding and distribution of drugs interacting with albumin and other transport proteins to influence the pharmacokinetics of many medications.
