Interpolated concentrations of native alpha 1 Acid Glycoprotein in human serum and plasma samples. The concentrations of alpha 1 Acid Glycoprotein were measured in duplicates, interpolated from the alpha 1 Acid Glycoprotein standard curves and corrected for sample dilution. Undiluted samples are as follows : serum 1 : 80,000, plasma (citrate) 40,000, plasma (EDTA) 1 : 80,000 and plasma (heparin) 1 : 80,000. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean alpha 1 Acid Glycoprotein concentration was determined to be 821.2 µg/mL in neat serum, 479.5 µg/mL in neat plasma (citrate), 881.7 µg/mL in neat plasma (EDTA) and 767.8 µg/mL in neat plasma (heparin).

Serum from ten individual healthy human male donors was measured in duplicate. Interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean alpha 1 Acid Glycoprotein concentration was determined to be 777.1 µg/mL with a range of 518.6 – 1,117 µg/mL.

The concentrations of alpha 1 Acid Glycoprotein were measured in duplicates, interpolated from the alpha 1 Acid Glycoprotein standard curves and corrected for sample dilution. Undiluted samples are as follows : urine 1 : 800, milk 1 : 500, saliva 1 : 800 and HepG2 cell culture supernatant 3.5%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean alpha 1 Acid Glycoprotein concentration was determined to be 6.85 µg/mL in neat urine, 5.34 µg/mL in neat milk, 8.42 µg/mL in neat saliva and 0.23 µg/mL in neat HepG2 cell culture supernatant.

How the assay works
Human Alpha 1 Acid Glycoprotein ELISA Kit, Chemiluminescent (ab326079) SimpleStep Ignite™ ELISA employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step during a 30 min only incubation, significantly reducing assay time. By utilizing a chemiluminscent detection method this improves the sensitivity and dynamic range when compared to traditional colorimetric ELISAs.
