Inhibitor of serine proteases. Its primary target is elastase, but it also has a moderate affinity for plasmin and thrombin. Irreversibly inhibits trypsin, chymotrypsin and plasminogen activator. The aberrant form inhibits insulin-induced NO synthesis in platelets, decreases coagulation time and has proteolytic activity against insulin and plasmin. Short peptide from AAT. Reversible chymotrypsin inhibitor. It also inhibits elastase, but not trypsin. Its major physiological function is the protection of the lower respiratory tract against proteolytic destruction by human leukocyte elastase (HLE).

How the assay works
Human alpha 1 Antitrypsin SimpleStep ELISA® employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Assay Specificity
Human IgM, human IgA and human IgE were prepared at 10 ng/mL and 250 ng/mL in Sample Diluent NS and assayed for cross reactivity. No cross-reactivity was observed for IgM or IgE at either concentration with a mean OD deviation from background of -0.01. No cross-reactivity was observed for IgA at 10 ng/mL and only 0.4% cross-reactivity at 250 ng/mL of IgA.
Purified human IgG was assayed at 5 ng/mL in the presence and absence of 250 ng/mL of human IgM, human IgA and human IgE to determine interference. After background subtraction, recovery of human IgG was observed at a mean of 98% with a standard deviation of 0.06.

alpha 1 Antitrypsin (SERPINA1) is a serine protease inhibitor found in a variety of biological fluids. While it can inhibit trypsin, chymotrypsin, and plasminogen activator, it's main target is elastase. Alpha 1 antitrypsin plays a critical role in the lung by inhibiting the activity of elastase, produced by neutrophils during inflammatory responses. Interpolated concentration of native alpha 1 Antitrypsin was measured in duplicate at different sample concentrations in 96-well vs. 384-well plates. Undiluted samples are 1:2,500 serum. The interpolated dilution factor corrected values (to neat) are plotted (mean +/- SD, n=2). Sample dilutions are made in Sample Diluent NS.
Serum of eight individual healthy human female donors was measured in duplicate. Interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean alpha 1 Antitrypsin concentration was determined to be 0.73 mg/mL with a range of 0.22 – 2.39 mg/mL.
Interpolated concentration of native alpha 1 Antitrypsin was measured in duplicate at different sample concentrations. Undiluted samples are as follows: serum 1:4,000, plasma (citrate)1:2,000, plasma (EDTA) 1:8,000, and plasma (heparin) 1:8,000.
