Interpolated concentrations of native Alpha-synuclein in human serum and plasmas samples.
The concentrations of Alpha-synuclein were measured in duplicates, interpolated from the Alpha-synuclein standard curves and corrected for sample dilution. Undiluted samples are as follows : serum 100%, plasma (EDTA) 100%, plasma (citrate) 50% and plasma (heparin) 100%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Alpha-synuclein concentration was determined to be 6.6 ng/mL in serum, 6.8 ng/mL in plasma (EDTA), 13.6 ng/mL in plasma (citrate), and 7.1 ng/mL in plasma (heparin).

Human Alpha-synuclein ELISA Kit ab260052 is a rapid single-wash 90-min ELISA kit to measure Human Alpha-synuclein in Tissue lysate, cell lysate, cell culture media, cell culture supernatant, plasma and serum. This SimpleStep ELISA® sensitivity is 504 pg/mL.
How the assay works
Human Alpha-synuclein SimpleStep ELISA® employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Assay Specificity
This kit recognizes both native and recombinant human human α-Synuclein protein in serum, plasma, and cell culture supernatant, cell and tissue extract samples only. Urine, saliva, and cerebrospinal fluid samples have not been tested with this kit. Both monomeric and aggregated Alpha-synuclein can be detected with this kit.

1. Mix: add samples/standards to the wells together with the capture and detector antibody cocktail. Incubate for 1 hr at room temperature
2. Wash
3. Add TMB development solution - incubate for 10 min

Biological function summary
The alpha-synuclein protein plays critical roles in neuronal activity. It contributes to neurotransmitter release regulation by acting in the formation and plasticity of the presynaptic neuronal network. Alpha-synuclein doesn't usually form parts of large protein complexes but it may associate transiently with membranes and vesicular structures. The protein's monomer form has also been observed in alpha lines and related neuronal processes operating alongside various cellular functions.
Pathways
Synaptic vesicle trafficking and dopamine neurotransmitter release are significant areas involving the alpha-synuclein protein. In these pathways alpha-synuclein interacts with other proteins like synaptophysin and protein monomer monomerizations are intrinsic to these processes. Altered function or aggregation of alpha-synuclein disrupts these pathways influencing broader neurological functions.
