Sandwich ELISA - Human ALT ELISA Kit (AB234578)
Interpolated concentration of native ALT was measured in duplicate at different sample concentrations in 96-well vs. 384-well plates. Undiluted samples are 25% serum. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). Sample dilutions are made in Sample Diluent NS + 1X Enhancer.

Interpolated concentrations of native ALT in human serum, plasma (EDTA) and plasma (citrate) samples.
The concentrations of ALT were measured in duplicates, interpolated from the ALT standard curves and corrected for sample dilution. Undiluted samples are as follows : serum 25%, plasma (citrate) 25%, and plasma (EDTA) 25%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean ALT concentration was determined to be 4,002.9 pg/mL in serum, 2,685.5 pg/mL in plasma (EDTA) and 1,886.6 pg/mL in plasma (citrate).

Linearity of dilution is determined based on interpolated values from the standard curve. Linearity of dilution defines a sample concentration interval in which interpolated target concentrations are directly proportional to sample dilution.
Native ALT was measured in the following biological samples in a 2-fold dilution series. Sample dilutions are made in Sample Diluent NS + 1X Enhancer.

Product details
Human ALT ELISA Kit ab234578 is a rapid single-wash 90-min ELISA kit to measure Human ALT in . This SimpleStep ELISA® sensitivity is 47.34 pg/mL.
How the assay works
Human ALT SimpleStep ELISA® employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details.
Assay Specificity
This kit recognizes both native and recombinant human ALT protein in serum, plasma, and cell culture supernatant samples only. Tissue extract samples have not been tested with this kit. This kit is incompatible with plasma- heparin and cell extract samples.
Human ALT ELISA Kit ab234578 protocol summary:
1. Mix: add samples/standards to the wells together with the capture and detector antibody cocktail. Incubate for 1 hr at room temperature
2. Wash
3. Add TMB development solution - incubate for 10 min
4. Add stop solution
5. Read the results on a plate reader at 450nm

Biological function summary
Alanine transaminase plays a critical role in amino acid metabolism and energy production. It facilitates the transamination process which is significant for the balance of amino acids and carbohydrates. ALT as a standalone enzyme does not form part of a protein complex but works closely with alanine aminotransferase reactions. It helps maintain the cellular nitrogen balance linking nitrogen metabolism with energy pathways.
Pathways
The role of alanine transaminase is to interconnect carbohydrate and amino acid metabolism. It participates in the gluconeogenesis pathway and the citric acid cycle important for energy production. Through its activity in these pathways it interacts with proteins like aspartate aminotransferase (AST) affecting the balance of amino and keto acids in cells.
