Interpolated concentrations of human AMH in serum, plasma (citrate), plasma (EDTA), and plasma (heparin). Interpolated concentration of native AMH was measured in duplicate at different sample concentrations. Undiluted samples are 45% serum, plasma (citrate), plasma (EDTA), and plasma (heparin).  The interpolated dilution factor corrected values (to neat sample) are plotted (mean +/- SD, n=2). Sample dilutions are made in Sample Diluent NS + 1X Enhancer.

Interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean AMH concentration of detected samples was determined to be 2,090 pg/mL with a range of undetected – 6,208 pg/mL.

How the assay works

Human AMH ELISA Kit, Chemiluminescent (ab326752) SimpleStep Ignite™ ELISA employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step during a 30 min only incubation, significantly reducing assay time. By utilizing a chemiluminscent detection method this improves the sensitivity and dynamic range when compared to traditional colorimetric ELISAs.

The anti-Muellerian hormone (AMH) plays an important role in several reproductive functions (PubMed : 14742691, PubMed : 34155118, PubMed : 3754790, PubMed : 8469238). Anti-Muellerian hormone binds and activates AMHR2, its specific type-II receptor, that heterodimerizes with type-I receptors (ACVR1 and BMPR1A) to regulate target gene expression through downstream SMAD protein signal transduction (PubMed : 20861221, PubMed : 34155118). Produced and secreted by Sertoli cells of the male fetus, anti-Muellerian hormone induces Muellerian duct regression during male fetal sexual differentiation (PubMed : 34155118, PubMed : 3754790, PubMed : 8469238). In female, it is produced by granulosa cells of the preantral and small antral follicles and acts as a negative regulator of the primordial to primary follicle transition and decreases FSH sensitivity of growing follicles (PubMed : 14742691). Also plays a role in Leydig cell differentiation and function (By similarity).
