Human Amyloid beta 1-40 ELISA Kit ab193692 is a sandwich ELISA to measure Human Amyloid beta 1-40 in serum, plasma, cell culture supernatant with a sensitivity of 0.1 ng/ml.

How the assay works

This assay employs an antibody specific for Human Amyloid beta 1-40 coated on a 96-well plate. Standards and samples are pipetted into the wells and the immobilized antibody captures Amyloid beta 1-40 present in the samples. The wells are washed and biotinylated anti-Human Amyloid beta 1-40 antibody is added. After washing away any unbound biotinylated antibody, an HRP-conjugated streptavidin is pipetted to the wells. After incubation, the wells are again washed, followed by the addition of a TMB substrate solution to the wells. Color will develop in proportion to the amount of Amyloid beta 1-40 bound in each well. Addition of the Stop Solution will change the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Amyloid beta 1-40 often referred to as beta-amyloid or Aβ40 is a peptide with a mass of approximately 4 kDa. It consists of 40 amino acids and constitutes a fragment cleaved from the amyloid precursor protein (APP). This peptide is expressed in various tissues primarily in the brain. Aβ40 is produced through enzymatic processing by β- and γ-secretases with β-APP40 ELISA kits commonly used to measure its levels in research.
Biological function summary

Amyloid beta 1-40 plays a role in cellular processes related to neuronal function and synaptic regulation. It does not act alone but often associates with other molecules forming oligomeric complexes. These complexes can influence membrane permeability and ionic homeostasis in neuronal cells. The peptide is known to disrupt cellular communication and in some contexts can lead to cellular toxicity when overproduced or misregulated.

Pathways

The amyloid beta 1-40 peptide is integral to the amyloidogenic pathway which is part of broader protein processing pathways in the brain. In this pathway it is closely tied to APP and interacts with presenilins which are components of the γ-secretase complex. Another significant pathway is linked to its cytotoxicity effects; this involves Tau protein where Aβ40 can influence hyperphosphorylation of Tau further impacting neurodegenerative processes.

Amyloid beta 1-40 is critically involved in Alzheimer's disease and is a hallmark of Alzheimer's pathology due to its aggregation into plaques. The peptide's accumulation correlates with neurodegenerative changes. Besides Alzheimer's Aβ40 has links to cerebral amyloid angiopathy a condition involving amyloid deposition in the walls of the brain's blood vessels. These diseases involve interactions between Aβ40 Tau and APOE proteins which are associated with disease progression and severity.
