Apolipoprotein CII also known as ApoC-II is an essential player in lipid metabolism. It is a 79 amino acid protein with an approximate mass of 8.9 kDa. ApoC-II is primarily expressed in the liver and secreted into the bloodstream where it associates with very-low-density lipoproteins (VLDLs) and chylomicrons. This protein serves an important mechanical role as an activator of lipoprotein lipase (LPL) an enzyme that hydrolyzes triglycerides into free fatty acids facilitating their uptake by tissues for energy production or storage.
Biological function summary

ApoC-II plays a critical role in the metabolism of plasma triglycerides. As an important component of the lipoprotein complex it is necessary for effective lipid transport and triglyceride regulation. By activating lipoprotein lipase ApoC-II ensures efficient breakdown of triglycerides in the bloodstream aiding in maintaining lipid homeostasis. Without ApoC-II LPL lacks full activity leading to impaired triglyceride clearance and dyslipidemia.

Pathways

ApoC-II is integral to lipid metabolic pathways particularly in the catabolism of triglyceride-rich lipoproteins. It participates significantly in the lipid digestion pathway where it functions in concert with proteins like ApoE and ApoB which further assist in lipid binding and transport. ApoC-II's regulatory actions within these pathways are important for preventing the build-up of triglycerides in circulation contributing to a balance in energy storage and metabolism.

Mutations or deficiencies in ApoC-II function can lead to hyperlipoproteinemia type Ib a condition characterized by elevated plasma triglycerides and risk of pancreatitis. This disorder manifests due to defective activation of lipoprotein lipase highlighting the target's role alongside proteins such as LPL in lipid clearance. Additionally ApoC-II interactions with cholesterol transport proteins may influence atherosclerosis a condition where plaque builds up in arteries posing further cardiovascular risks.

Product protocols

Target data

Component of chylomicrons, very low-density lipoproteins (VLDL), low-density lipoproteins (LDL), and high-density lipoproteins (HDL) in plasma. Plays an important role in lipoprotein metabolism as an activator of lipoprotein lipase. Both proapolipoprotein C-II and apolipoprotein C-II can activate lipoprotein lipase. In normolipidemic individuals, it is mainly distributed in the HDL, whereas in hypertriglyceridemic individuals, predominantly found in the VLDL and LDL.
