Product details

Abcam's Apolipoprotein H (APOH) Human in vitro ELISA (Enzyme-Linked Immunosorbent Assay) kit is designed for the quantitative measurement of Apolipoprotein H concentrations in Human plasma, serum, saliva, urine, milk and cell culture samples.

An Apolipoprotein H specific antibody has been precoated onto 96-well plates and blocked. Standards or test samples are added to the wells and subsequently an Apolipoprotein H specific biotinylated detection antibody is added and then followed by washing with wash buffer. Streptavidin-Peroxidase Complex is added and unbound conjugates are washed away with wash buffer. TMB is then used to visualize Streptavidin-Peroxidase enzymatic reaction. TMB is catalyzed by Streptavidin-Peroxidase to produce a blue color product that changes into yellow after adding acidic stop solution. The density of yellow coloration is directly proportional to the amount of Apolipoprotein H captured in plate.

The entire kit may be stored at -20°C for long term storage before reconstitution - Avoid repeated freeze-thaw cycles.

Supplementary information

Apolipoprotein H also known as Apo H or β2-glycoprotein I is a plasma glycoprotein with a molecular weight of approximately 50 kDa. It is expressed predominantly in the liver and is found circulating in the bloodstream. Apo H plays a mechanical role in binding to negatively charged substances such as phospholipids on cell membranes. This binding capability allows it to modulate the immune response and participate in lipid metabolism.
Biological function summary

This protein interacts with various components in the blood contributing to the modulation of blood coagulation and immune function. Apolipoprotein H exists as a single polypeptide chain and does not form a complex in its plasma state but it can associate with lipoproteins. It functions as an important co-factor in the inhibition of the coagulation pathway and also affects platelet aggregation. Both of these activities make Apo H important in preventing undesirable thrombosis.

Pathways

Apo H plays a role in lipid transport and metabolic pathways. It is particularly involved in the regulation of the coagulation cascade linking with other proteins such as high-density lipoprotein (HDL) particles and possibly impacting lipoprotein clearance. By interacting with phospholipids and apolipoproteins Apo H aids in smooth flow and regulation of these biological pathways.

Apo H shows associations with autoimmune disorders like antiphospholipid syndrome a condition characterized by recurrent thrombosis and pregnancy complications. Additionally research suggests a connection to cardiovascular diseases through its interaction with proteins like oxidized LDL. These interactions make it a focus in the study of disorders related to thrombosis and cardiovascular health.

Target data

Binds to various kinds of negatively charged substances such as heparin, phospholipids, and dextran sulfate. May prevent activation of the intrinsic blood coagulation cascade by binding to phospholipids on the surface of damaged cells.
