Function

Mediates inactivation of the lipoprotein lipase LPL, and thereby plays a role in the regulation of triglyceride clearance from the blood serum and in lipid metabolism (PubMed:19270337, PubMed:21398697, PubMed:27929370, PubMed:29899144). May also play a role in regulating glucose homeostasis and insulin sensitivity (Probable). Inhibits proliferation, migration, and tubule formation of endothelial cells and reduces vascular leakage (PubMed:14583458, PubMed:17068295). Upon heterologous expression, inhibits the adhesion of endothelial cell to the extracellular matrix (ECM), and inhibits the reorganization of the actin cytoskeleton, formation of actin stress fibers and focal adhesions in endothelial cells that have adhered to ANGPTL4-containing ECM (in vitro) (PubMed:17068295). Depending on context, may modulate tumor-related angiogenesis (By similarity).ANGPTL4 N-terminal chainMediates inactivation of the lipoprotein lipase LPL, and thereby plays an important role in the regulation of triglyceride clearance from the blood serum and in lipid metabolism (PubMed:19270337, PubMed:21398697, PubMed:27929370, PubMed:29899144). Has higher activity in LPL inactivation than the uncleaved protein (PubMed:19270337, PubMed:21398697).

Notes

Abcam's ARP4 (Angiopoietin-like 4) Human ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Human ARP4 in serum, plasma, and cell culture supernatants.

This assay employs an antibody specific for Human ARP4 coated on a 96-well plate. Standards and samples are pipetted into the wells and ARP4 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Human ARP4 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of ARP4 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Optimisation may be required with urine samples.

Supplementary info

Activity summary

The ARP4 protein also known as Actin-Related Protein 4 plays a role in chromatin remodeling which is important for DNA-related processes like transcription replication and repair. ARP4 has a molecular mass of approximately 50 kDa and is ubiquitously expressed in eukaryotic cells. It remains localized mainly within the nucleus where it contributes to changes in the configuration of chromatin structure permitting the necessary access of various transcription machinery to DNA.

Biological function summary

ARP4 functions as an integral component of the INO80 chromatin remodeling complex. This complex is essential for modifying chromatin structure which impacts gene expression regulation and DNA repair. Through its association with the INO80 complex ARP4 contributes to the mobilization and spacing of nucleosomes important for maintaining genomic stability. It also further enhances DNA repair processes by allowing more accessible interactions with DNA sites requiring repair.

Pathways

ARP4 is involved in the regulation of gene expression and DNA damage response. The protein plays a part in the chromatin remodeling pathway and the DNA repair pathway. Through its work with the INO80 complex ARP4 associates with other remodelers like RUVBL1 and RUVBL2 which are also components of the same complex. These interactions facilitate effective transcriptional regulation and the DNA damage response showcasing the critical nature of ARP4 in maintaining normal cellular functions.

Associated diseases and disorders

ARP4 has a place in connecting with conditions like cancer and neurodegenerative diseases. Aberrant expression or mutations in ARP4 and its associated complexes can disrupt chromatin structure leading to misregulation of gene expression which can precipitate tumorigenesis or neuronal damage. Through its impact on chromatin remodeling and gene regulation ARP4 works alongside other proteins like BRG1 a part of the SWI/SNF chromatin remodeling complex influencing pathways that may result in these diseases.
