Abcam's ARP4 (Angiopoietin-like 4) Human ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Human ARP4 in serum, plasma, and cell culture supernatants.
This assay employs an antibody specific for Human ARP4 coated on a 96-well plate. Standards and samples are pipetted into the wells and ARP4 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Human ARP4 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of ARP4 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.
Optimisation may be required with urine samples.

Biological function summary
ARP4 functions as an integral component of the INO80 chromatin remodeling complex. This complex is essential for modifying chromatin structure which impacts gene expression regulation and DNA repair. Through its association with the INO80 complex ARP4 contributes to the mobilization and spacing of nucleosomes important for maintaining genomic stability. It also further enhances DNA repair processes by allowing more accessible interactions with DNA sites requiring repair.
Pathways
ARP4 is involved in the regulation of gene expression and DNA damage response. The protein plays a part in the chromatin remodeling pathway and the DNA repair pathway. Through its work with the INO80 complex ARP4 associates with other remodelers like RUVBL1 and RUVBL2 which are also components of the same complex. These interactions facilitate effective transcriptional regulation and the DNA damage response showcasing the critical nature of ARP4 in maintaining normal cellular functions.
