Responsible for the proteolytic processing of the amyloid precursor protein (APP). Cleaves at the N-terminus of the A-beta peptide sequence, between residues 671 and 672 of APP, leads to the generation and extracellular release of beta-cleaved soluble APP, and a corresponding cell-associated C-terminal fragment which is later released by gamma-secretase (PubMed:10656250, PubMed:10677483, PubMed:20354142). Cleaves CHL1 (By similarity).

This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Interpolated concentration of native BACE1 was measured in duplicate at different sample concentrations. Undiluted samples are 50% cerebrospinal fluid. The interpolated dilution factor corrected values (to neat sample) are plotted (mean +/- SD, n=2). Sample dilutions are made in Sample Diluent NS.
Interpolated concentration of native BACE1 was measured in duplicate at different sample concentrations in 96-well vs. 384-well plates. Undiluted samples are 125 µg/mL human cortex tissue extract. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). Sample dilutions are made in Sample Diluent NS + 1X Enhancer.

Serum of seven individual healthy human female donors was measured in duplicate. Interpolated dilution factor corrected values (to neat sample) are plotted (mean +/- SD, n=2). The mean BACE1 concentration was determined to be 1.26 ng/mL with a range of 0.62 – 2.11 ng/mL.
