Other species reactivity was determined by measuring 150 ug/mL extract load of human and mouse heart tissue extracts and interpolating the protein concentrations from the BAG3 standard curve. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). Concentrations are expressed in ng of protein per mg of extract. The mean Bag3 concentration was determined to be 18.15 ng/mg in human heart tissue extract and 14.86 ng/mg in mouse heart tissue extract.

Interpolated concentrations of native BAG3 in HEK293T cell extract based on a 100 µg/mL extract load and human heart tissue extract based on a 150 µg/mL extract load. The concentrations of BAG3 were measured in duplicate and interpolated from the BAG3 standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean BAG3 concentration was determined to be 2,497 pg/mL in HEK293T cell extract and 2,743 pg/mL in human heart tissue extract.

How the assay works

Human BAG3 ELISA Kit, Chemiluminescent (ab325726) SimpleStep Ignite™ ELISA employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step during a 30 min only incubation, significantly reducing assay time. By utilizing a chemiluminscent detection method this improves the sensitivity and dynamic range when compared to traditional colorimetric ELISAs.

Target data

Co-chaperone and adapter protein that connects different classes of molecular chaperones including heat shock proteins 70 (HSP70s), e.g. HSPA1A/HSP70 or HSPA8/HSC70, and small heat shock proteins (sHSPs), e.g. HSPB8 (PubMed : 27884606, PubMed : 30559338). Acts as a nucleotide-exchange factor (NEF) promoting the release of ADP from HSP70s, thereby triggering client protein release (PubMed : 27884606, PubMed : 30559338). Nucleotide release is mediated via BAG3 binding to the nucleotide-binding domain (NBD) of HSP70s, whereas client release is mediated via binding to the substrate-binding domain (SBD) (PubMed : 27474739, PubMed : 9873016). Has anti-apoptotic activity (PubMed : 10597216). Plays a role in the HSF1 nucleocytoplasmic transport (PubMed : 26159920).
