Sandwich ELISA - Human Beclin-1 ELISA Kit (AB254511)
Example of human Beclin-1 standard curve in 1X Cell Extraction Buffer PTR.
The Beclin-1 standard curve was prepared as described in Section 10. Raw data values are shown in the table. Background-subtracted data values (mean +/- SD) are graphed.

Sandwich ELISA - Human Beclin-1 ELISA Kit (AB254511)
Interpolated concentrations of native Beclin-1 in human MCF-7 cell extract based on a 1,000 μg/mL extract load.
The concentrations of Beclin-1 were measured in duplicate and interpolated from the Beclin-1 standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Beclin-1 concentration was determined to be 10.03 ng/mL.

Product details
Human Beclin-1 ELISA Kit ab254511 is a rapid single-wash 90-min Sandwich ELISA to measure Human Beclin-1 in cell culture extracts. This SimpleStep sensitivity is 671 pg/mL.
How the assay works
Human Beclin-1 SimpleStep ELISA®employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details.
Assay Specificity
Our SimpleStep ELISA® kits use recombinant monoclonal antibodies rigorously validated to ensure the highest level of consistency and reproducibility, improved sensitivity and specificity and ease of scalability and security of supply.
Please refer to our protocol booklet for more details.

We offer the antibody pair used in this kit in a BSA and Azide-free format, ready for conjugation:
- Anti-Beclin 1 antibody [EPR20473-245] - BSA and Azide free (Capture) ab252668
- Anti-Beclin 1 antibody [EPR20473-221] - BSA and Azide free (Detector) ab252668
Beclin-1, also known as BECN1, plays a key role in the regulation of autophagy and cell death. Together with PI3KC3, Beclin-1 nucleates autophagosome formation and is essential for proper degradation of cellular components. Improper beclin-1 signaling is associated with schizophrenia, tumorigenesis, and neurodegeneration.

Biological function summary
The protein functions as an important component of the Beclin 1 autophagy complex which orchestrates the autophagic process. This complex is essential for promoting autophagosome nucleation by interacting with multiple partners that regulate the autophagy pathway. In addition to its role in autophagy Beclin 1 protein stabilizes interactions with other autophagic proteins ensuring the correct assembly and activity of this cellular mechanism. The regulation of Beclin 1's activity and expression influences the efficiency and specificity of autophagy in cells.
Pathways
Beclin 1 integrates into critical cellular pathways like the PI3K/Akt pathway and the mTOR pathway which are pivotal in cell growth and survival. Within these pathways beclin cooperates closely with proteins such as ATG14 and VPS34. These interactions facilitate the recruitment and activation of additional factors that promote the elongation of phagophores which eventually become complete autophagosomes. The coordination of these pathways ensures the proper balance between cell survival and death adapting to cellular conditions.
