Interpolated concentrations of native Human beta-Catenin in HeLa cell extract HEK293 cell extract A431 cell extract and A549 cell extract based on a 400 µg/mL extract load.

The concentrations of Human beta-Catenin were measured in duplicate and interpolated from the Human beta-Catenin standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD n=2). The mean Human beta-Catenin concentration was determined to be 45.8 ng/mL in HeLa 58.5 ng/mL in HEK293 215 ng/mL in A431 and 49 ng/mL in A549.

How the assay works

Human Beta Catenin ELISA Kit, Chemiluminescent (ab327292) SimpleStep Ignite™ ELISA employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step during a 30 min only incubation, significantly reducing assay time. By utilizing a chemiluminscent detection method this improves the sensitivity and dynamic range when compared to traditional colorimetric ELISAs.

In the absence of Wnt, forms a complex with AXIN1, AXIN2, APC, CSNK1A1 and GSK3B that promotes phosphorylation on N-terminal Ser and Thr residues and ubiquitination of CTNNB1 via BTRC and its subsequent degradation by the proteasome (PubMed : 17524503, PubMed : 18077326, PubMed : 18086858, PubMed : 18957423, PubMed : 21262353, PubMed : 22155184, PubMed : 22647378, PubMed : 22699938).
