Species reactivity was determined by measuring 100 ug/mL extract of human HeLa, mouse NIH-3T3 and rat PC12 cell extracts and interpolating the protein concentrations from the Human standard curve.
The mean BMI1 concentration was determined to be 16.9 ng/mg in human HeLa extract, 12.8 ng/mg in mouse NIH-3T3 extract and 1.2 ng/mg in rat PC12 extract.

Interpolated concentrations of native BMI1 in human HeLa and K562 based on 300 μg/mL and 150 µg/mL extract loads, respectively.
The concentrations of BMI1 were measured in duplicate and interpolated from the BMI1 standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean BMI1 concentration was determined to be 5,142 pg/mL in HeLa extract and 3,399 pg/mL in K562 extract.

Human BMI1 ELISA kit (ab272210) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of BMI1 protein in cell and tissue extracts. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human BMI1 with 34 pg/mL sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:

BMI1 (Polycomb Complex Protein BMI-1), also known as Polycomb group RING Finger Protein 4, is a component of a Polycomb group (PcG) multiprotein PRC1-like complex, a complex class required to maintain the transcriptionally repressive state of many genes, including Hox genes, throughout development. PcG PRC1 complex acts via chromatin remodeling and modification of histones; it mediates monoubiquitination of histone H2A 'Lys-119', rendering chromatin heritably changed in its expressibility. The complex composed of RNF2, UB2D3 and BMI1 binds nucleosomes, and has activity only with nucleosomal histone H2A. In the PRC1-like complex, BMI1 regulates the E3 ubiquitin-protein ligase activity of RNF2/RING2. Human BMI1 amino acids sequence is 97% identical to mouse BMI1 and is 95% identical to rat BMI1.

Biological function summary
The functions of Bmi1 extend into maintaining stem cell renewal and differentiation. It acts as part of the Polycomb repressive complex 1 (PRC1) partnering with other proteins to regulate gene expression by altering chromatin structure. Bmi1 protein regulates cell proliferation and inhibits senescence by repressing the INK4a/ARF locus which encodes tumor suppressor proteins like p16INK4a and p14ARF.
Pathways
Bmi1 plays significant roles in processes such as the DNA damage response and the self-renewal of hematopoietic stem cells. It is involved in the modulation of the Wnt and Notch signaling pathways. Bmi1 protein interacts closely with proteins such as CBX2 and RNF2 within these pathways affecting the transcriptional landscape that governs cell fate decisions.
