Growth factor of the TGF-beta superfamily that plays essential roles in many developmental processes, including cardiogenesis, neurogenesis, and osteogenesis (PubMed:18436533, PubMed:24362451, PubMed:31019025). Induces cartilage and bone formation (PubMed:3201241). Initiates the canonical BMP signaling cascade by associating with type I receptor BMPR1A and type II receptor BMPR2 (PubMed:15064755, PubMed:17295905, PubMed:18436533). Once all three components are bound together in a complex at the cell surface, BMPR2 phosphorylates and activates BMPR1A (PubMed:7791754). In turn, BMPR1A propagates signal by phosphorylating SMAD1/5/8 that travel to the nucleus and act as activators and repressors of transcription of target genes. Also acts to promote expression of HAMP, via the interaction with its receptor BMPR1A/ALK3 (PubMed:31800957). Can also signal through non-canonical pathways such as ERK/MAP kinase signaling cascade that regulates osteoblast differentiation (PubMed:16771708, PubMed:20851880). Also stimulates the differentiation of myoblasts into osteoblasts via the EIF2AK3-EIF2A-ATF4 pathway by stimulating EIF2A phosphorylation which leads to increased expression of ATF4 which plays a central role in osteoblast differentiation (PubMed:24362451). Acts as a positive regulator of odontoblast differentiation during mesenchymal tooth germ formation, expression is repressed during the bell stage by MSX1-mediated inhibition of CTNNB1 signaling (By similarity).

Human BMP2 ELISA kit (ab277085) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Human BMP2 protein in human serum, plasma, and cell culture media. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human BMP2 with 79.8 pg/mL sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Bone morphogenetic protein-2 (BMP2, BMP 2A) belongs to the transforming growth factor beta (TGF-β) superfamily. It is thought to be involved in cartilage and bone formation during embryogenesis, but may have additional functions in morphogenesis as implied by its expression in various organs and embryonic tissues of mice. BMP2 has been identified as a candidate mediator of retinoid activity. BMP2 protein induces medulloblastoma cell apoptosis, whereas the BMP2 antagonist noggin blocks both retinoid and BMP2-induced apoptosis. BMP2 also induces p38 mitogen-activated protein kinase (MAPK), which is necessary for BMP2 and retinoid-induced apoptosis. Bone morphogenic proteins (BMPs) are known to promote osteogenesis, and clinical trials are currently underway to evaluate the ability of certain BMPs to promote fracture-healing and spinal fusion.

The BMP2 standard curve was prepared as described in Section 10. Raw data values are shown in the table. Background-subtracted data values (mean +/- SD) are graphed.
Interpolated concentrations of spike BMP2 in human serum, plasma and cell culture media samples.
The concentrations of BMP2 were measured in duplicates, interpolated from the BMP2 standard curves and corrected for sample dilution. Undiluted samples are as follows: serum 25%, plasma (citrate) 25%, plasma (EDTA) 25%, plasma (heparin) 25% and cell culture media 25%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2).
