Interpolated concentrations of native BMP9 in human serum, plasma and cell culture supernatant samples.
The concentrations of BMP9 were measured in duplicates, interpolated from the BMP9 standard curves and corrected for sample dilution. Undiluted samples are as follows : serum 100%, plasma (citrate) 50%, and HepG2 cell culture supernatant 100%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean BMP9 concentration was determined to be 213.4 pg/mL in serum, 166.3 pg/mL in plasma (citrate) and 37.3 pg/mL in HepG2 cell culture supernatant.

How the assay works
Human BMP9 SimpleStep ELISA®employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. 1. Mix: add samples/standards to the wells together with the capture and detector antibody cocktail. Incubate 1 hr at room temperature
2. Wash
3. Add TMB development solution - incubate for 10 min
4. Add Stop solution
5. Read the results on a plate reader at 450 nm

Bone morphogenetic protein 9 (BMP9), also known as growth differentiation factor 2, is a protein encoded by the GDF2 gene and expressed in the liver. BMP9 is a member of the TGF-? superfamily that plays a role in angiogenesis as a potent inhibitor as well as a potent regulator for osteogenesis, glucose metabolism, and vascular tone. Studies have shown that mutations in BMP9 cause vascular disorders. Mouse, rat, cow, monkey share 96.4%, 94.5%, 92.7%, and 97.3% sequence homology, respectively.
