Interpolated concentrations of native Bone Sialoprotein in human serum and plasma samples.

The concentrations of Bone Sialoprotein were measured in duplicates, interpolated from the Bone Sialoprotein standard curves and corrected for sample dilution. Undiluted samples are as follows : serum 100%, plasma (citrate) 100%, plasma (heparin) 100% and plasma (EDTA) 100%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Bone Sialoprotein concentration was determined to be 471 pg/mL in serum, 1,397 pg/mL in plasma (citrate) and 647 pg/mL in plasma (heparin) and 434 pg/mL in plasma (EDTA).

Product details

Human Bone Sialoprotein ELISA Kit ab277706 is a rapid single-wash 90-min Sandwich ELISA to measure Human Bone Sialoprotein in citrate plasma, EDTA plasma, heparin plasma, serum. This SimpleStep sensitivity is 9 pg/mL.

How the assay works

Human Bone Sialoprotein SimpleStep ELISA®employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details.

Assay Specificity

Our SimpleStep ELISA® kits use recombinant monoclonal antibodies rigorously validated to ensure the highest level of consistency and reproducibility, improved sensitivity and specificity and ease of scalability and security of supply.
Please refer to our protocol booklet for more details.

Human Bone Sialoprotein ELISA Kit ab277706 protocol summary

1. Mix: add samples/standards to the wells together with the capture and detector antibody cocktail. Incubate 1 hr at room temperature
2. Wash
3. Add TMB development solution - incubate for 10 min
4. Add Stop solution

Design your own immunoassay

We offer the antibody pair used in this kit in a BSA and Azide-free format, ready for conjugation:

- Anti-Bone Sialoprotein antibody [EPR23243-85] - BSA and Azide free (Capture) ab277998
- Anti-Bone Sialoprotein antibody [EPR23243-13] - BSA and Azide free (Detector) ab277998

Bone Sialoprotein, also known as IBSP, is a matrix protein that plays a role in mineralization of bone and other calcified structures. The exact function of this protein is currently under investigation. The immunogen with which the antibodies in this kit were raised is composed of the full-length mature protein. This protein is not homologous with other species; as such, it is not expected to cross-react.

Supplementary information

Bone Sialoprotein (BSP) is a non-collagenous glycoprotein of the SIBLING (Small Integrin-Binding Ligand N-linked Glycoprotein) family with a mass around 70-80 kDa. Its expression mainly takes place in mineralized tissues like bone and dentin highlighting its importance in skeletal biology. BSP has a strong affinity for calcium and hydroxyapatite allowing it to play binding and structural roles in bone matrix formation.
Biological function summary

The protein influences the nucleation of hydroxyapatite crystals and helps regulate mineralization processes. It associates with collagenous and non-collagenous components in the bone and contributes to tissue integrity and repair. BSP functions as part of a complex matrix that orchestrates cell adhesion and signal transduction impacting osteoblast and osteoclast activity.

Pathways

Bone Sialoprotein interacts within the extracellular matrix organization pathway and the integrin signaling pathway. In the extracellular matrix pathway BSP plays a role together with proteins like osteopontin also from the SIBLING family to influence cellular adhesion. Through integrin signaling BSP modulates cell-surface interactions particularly affecting the behavior of osteoclasts and osteoblasts and their capacity to maintain bone homeostasis.

Bone Sialoprotein associates with pathological conditions such as osteosarcoma and osteoporosis. In osteosarcoma BSP overexpression correlates with aggressive tumor growth potentially linking it with the tumor protein osteopontin suggesting a marker for malignancy. In osteoporosis dysregulation of BSP expression may affect bone density contributing to weakened bone structure alongside changes in other bone matrix proteins.

Target data

Binds tightly to hydroxyapatite (PubMed : 11459848). Appears to form an integral part of the mineralized matrix (PubMed : 1818768). Probably important to cell-matrix interaction (PubMed : 1818768). Promotes adhesion and migration of various cells via the alpha-V/beta-3 integrin receptor (ITGAV : ITGB3) (PubMed : 10640428, PubMed : 11459848, PubMed : 24103036).
