How the assay works
This assay employs an antibody specific for human C3a coated on a 96-well plate. Standards and samples are pipetted into the wells and C3a present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-human C3a antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of C3a bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm

C3b another key fragment binds to pathogen surfaces aiding opsonization and immune clearance.
Pathways
C3's function plays an important role in the alternative and classical complement pathways. In the alternative pathway C3b contributes directly by forming a C3 convertase complex. This interaction facilitates further cleavage of C3 amplifying immune response. C3b also associates with proteins like Factor B and Factor D to stabilize the convertase complex and enhance opsonization. In the classical pathway C3 interacts with components like C4b to optimize pathogen elimination.
