Serum from eight individual healthy C8b female donors was measured in duplicate. Interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean C8b concentration was determined to be 57 μg/mL with a range of 31 – 120 μg/mL.

Interpolated concentrations of native C8b in human serum, plasma (EDTA), plasma (Citrate), and plasma (Heparin). The concentrations of C8b were measured in duplicates, interpolated from the target standard curves and corrected for sample dilution. Undiluted samples are as follows : serum 1 : 800, plasma (EDTA) 1 : 1,600, Plasma (Citrate) 1 : 800, plasma (Heparin) 1 : 400. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean target concentration was determined to be 16.3 μg/mL in serum, 34.3 μg/mL in plasma (EDTA), 12.1 μg/mL in plasma (Citrate), and 8.8 μg/mL in plasma (Heparin).

Human C8b (Complement C8b) (ab283548) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Human DystrophinH protein in human cell and tissue extract samples. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human C8b (Complement C8b) with 0.1 ng/mL sensitivity.

SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Complement component C8 beta chain is a protein encoded by the C8B gene. C8 is one of the components of the membrane attack complex (MAC) which mediates cell lysis and initiates membrane penetration of the complex. C8B contains TSP type-1 1, LDL-receptor class A, MACPF, EGF-like, and TSP type-1 2 domains. C8B is a 591 amino acid protein.

Complement factor 8 beta (C8B) is an essential protein component of the complement system an important part of the immune response. Commonly referred to as C8B this protein has a molecular mass of approximately 64 kilodaltons. C8B is primarily expressed in liver tissue where it contributes to the formation of the C8 protein complex a component necessary for the assembly of the membrane attack complex (MAC). Other names for this target include complement component C8 beta chain emphasizing its role in the complement cascade.
Biological function summary

The C8B protein facilitates the stabilization of the C8 complex which also consists of the alpha and gamma chains to perform its role in the immune system. It plays an important role in forming the MAC an important structure for lysing pathogenic cells like bacteria by disrupting their plasma membranes. This action highlights the importance of C8B in innate immunity where it enables the host to eliminate harmful cells effectively.

Pathways

C8B's activities are integral to the complement cascade specifically within the terminal membrane attack pathway. It functions in conjunction with other complement factors like complement factor 5 to 9 forming a cascade of sequential activations. C8B works closely with complement factor 8 alpha and complement factor C5 within this pathway to achieve effective immune responses. C8B has significant connections to immunodeficiencies and inflammatory conditions. Patients with deficiencies in any of the components forming the MAC including C8B often suffer from recurrent Neisserial infections due to impaired bacterial lysis. C8B mutations contribute to properdin-independent pathway issues that can lead to susceptibility to such infections. Moreover malfunction in C8B can associate with autoimmune disorders implying a disruption in the delicate balance of the immune response where proteins like factor 8 and complement factor 5 play critical roles in maintaining immunity.

Component of the membrane attack complex (MAC), a multiprotein complex activated by the complement cascade, which inserts into a target cell membrane and forms a pore, leading to target cell membrane rupture and cell lysis (PubMed : 22832194, PubMed : 26841837, PubMed : 27052168, PubMed : 30552328, PubMed : 7440581). The MAC is initiated by proteolytic cleavage of C5 into complement C5b in response to the classical, alternative, lectin and GZMK complement pathways (PubMed : 30552328, PubMed : 39914456, PubMed : 39814882, PubMed : 7440581). The complement pathways consist in a cascade of proteins that leads to phagocytosis and breakdown of pathogens and signaling that strengthens the adaptive immune system (PubMed : 30552328, PubMed : 7440581). C8B, together with C8A and C8G, inserts into the target membrane, but does not form pores by itself (PubMed : 30552328). During MAC assembly, associates with C5b, C6 and C7 to form the C5b8 intermediate complex that inserts into the target membrane and traverses the bilayer increasing membrane rigidity (PubMed : 30552328, PubMed : 6833260).
