Human Cathepsin D (mature, single chain) ELISA Kit- Extracellular is a single-wash 90-min Simplestep used to quantify Human Cathepsin D (mature, single chain) with a sensitivity of 12.58 pg/mL. The assay uses a simple mix-wash-read protocol with just one incubation and one wash step.
- Colorimetric Sandwich ELISA - 450 nm readout : works on any standard plate reader
- Different formats for different needs: 10x96 plates for bulk orders and 384-well plate for higher throughput
- Design your own immunoassay: we also offer the conjugation-ready antibody pair

Acid protease active in intracellular protein breakdown. Plays a role in APP processing following cleavage and activation by ADAM30 which leads to APP degradation (PubMed:27333034). Involved in the pathogenesis of several diseases such as breast cancer and possibly Alzheimer disease.

Human Cathepsin D (mature, single chain) SimpleStep ELISA® kit is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Cathepsin D (mature, single chain) protein in Serum, Cit Plasma, Hep Plasma, Cell culture supernatant, Cell culture supernatant, Urine, Saliva, Milk. Quantitate Human Cathepsin D (mature, single chain) with 12.58 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:

Interpolated concentrations of human Cathepsin D in HepG2 supernatant in 96-well vs. 384-well plates.
Interpolated concentration of native Cathepsin D was measured in duplicate at different sample concentrations in 96-well vs. 384-well plates. Undiluted samples are 25% HepG2 supernatant. The interpolated dilution factor corrected values (to neat sample) are plotted (mean +/- SD, n=2). Sample dilutions are made in Sample Diluent NS + 1X Enhancer.

Example of human Cathepsin D standard curve in 96-well vs. 384-well plate. Background-subtracted data values (mean +/- SD) are graphed.
Interpolated concentrations of Cathepsin D in serum from donors with type 2 diabetes (T2D), compared to pooled normal human serum.
Serum of 3 individual human donors diagnosed with type 2 diabetes (T2D) was measured in duplicate. Interpolated dilution factor corrected values (to neat sample) are plotted (mean +/- SD, n=2). The mean Cathepsin D concentration of T2D samples was determined to be 1,975.95 pg/mL with a range of 1,314.15 – 2,776.70 pg/mL.
Interpolated concentrations of Cathepsin D in normal human serum donors.
Serum of 12 individual healthy human donors (6 female and 6 male) was measured in duplicate. Interpolated dilution factor corrected values (to neat sample) are plotted (mean +/- SD, n=2). The mean Cathepsin D concentration of detected samples was determined to be 213.08 pg/mL with a range of undetected – 386.16 pg/mL.
Interpolated concentrations of human Cathepsin D in cell culture supernatant and saliva.
Interpolated concentration of native Cathepsin D was measured in duplicate at different sample concentrations. Undiluted samples are as follows: A431 supernatant 6.25%, HepG2 supernatant 25%, and male saliva 25%. The interpolated dilution factor corrected values (to neat sample) are plotted (mean +/- SD, n=2). Sample dilutions are made in Sample Diluent NS + 1X Enhancer.
Interpolated concentrations of human Cathepsin D in serum and plasma.
Interpolated concentration of native Cathepsin D was measured in duplicate at different sample concentrations. Undiluted samples are 50% serum, plasma (citrate), and plasma (heparin). The interpolated dilution factor corrected values (to neat sample) are plotted (mean +/- SD, n=2). Sample dilutions are made in Sample Diluent NS + 1X Enhancer.
