Interpolated concentration of native CD163 was measured in duplicate at different sample concentrations in 96-well vs 384-well plates. Undiluted samples are 1 : 100 serum. The interpolated dilution factor corrected values (to neat sample) are plotted (mean +/- SD, n=2). Sample dilutions are made in Sample Diluent 25BS.

Interpolated concentrations of native CD163 in stimulated human PBMC cell culture supernatant samples.
PBMC samples were stimulated for 46 hours with 1.5% PHA-M. The concentrations of CD163 were measured in duplicates, interpolated from the CD163 standard curves and corrected for sample dilution. Undiluted samples are as follows : stimulated PBMC supernatant 25%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean CD163 concentration was determined to be 22 ng/mL in stimulated PBMC supernatant.

Interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean CD163 concentration was determined to be 563 ng/mL with a range of 338 – 1,030 ng/mL.

The concentrations of CD163 were measured in duplicates, interpolated from the CD163 standard curves and corrected for sample dilution. Undiluted samples are as follows : serum 1%, plasma (citrate) 1%, plasma (heparin) 1%, and plasma (EDTA) 1%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean CD163 concentration was determined to be 704 ng/mL in serum, 552 ng/mL in plasma (citrate), 545 ng/ml in plasma (heparin), and 515 ng/mL in plasma (EDTA).

How the assay works
Human CD163 SimpleStep ELISA®employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. CD163 is a scavenge receptor for the haptoglobin-hemoglobin complex. CD163 clears the complex in the liver, spleen and circulation. Soluble CD163 also plays a role in reducing inflammation, and dysregulation of CD163 is associated with some inflammatory conditions like multiple sclerosis, rheumatoid arthritis and diabetes mellitus.

Biological function summary
CD163 participates in anti-inflammatory processes and is linked to immune modulation. It plays a pivotal role in the resolution of inflammation and promotes the removal of damaged cells and tissues. Through its involvement in the endocytic pathway CD163 interacts with various ligands facilitating interactions with other proteins. In the context of research CD163 functionality can be analyzed using techniques like CD163 IHC CD163 ELISA and CD163 stain in both human and mouse models.
Pathways
CD163 is integral to the inflammation and immune response pathways. It participates in the heme-oxygenase pathway which is essential in the breakdown of heme into biliverdin free iron and carbon monoxide. This pathway involvement is tightly linked with heme oxygenase-1 (HO-1) a cytoprotective enzyme that helps to mitigate inflammatory responses. The interaction with haptoglobin and related proteins provides further insight into its regulatory roles within the immune system.
