2.5% Serum from ten individual healthy human female donors was measured in duplicate.
Interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean CD226 concentration was determined to be 9.36 ng/mL with a range of 2.62 – 25.51 ng/mL.

Interpolated concentrations of native CD226 in human serum, plasma, and cell culture supernatant samples.
The concentrations of CD226 were measured in duplicates, interpolated from the CD226 standard curves and corrected for sample dilution. Undiluted samples are as follows : serum 5%, plasma (citrate) 10%, plasma (EDTA) 10%, plasma (heparin) 2.5%, and PBMC supernatant 50%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean CD226 concentration was determined to be 5.77 ng/mL in serum, 3.72 ng/mL in plasma (citrate), 2.73 ng/mL in plasma (EDTA), 22.06 ng/mL in plasma (heparin), and 0.34 ng/mL in PBMC supernatant.

Human CD226 ELISA kit (ab282303) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Human CD226 protein in human serum, plasma, and cell culture supernatant samples. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human CD226 with 1.58 pg/mL sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Human CD226, a glycoprotein, is expressed on the surface of NK cells, platelets, monocytes, and a subset of T cells. CD226 is involved in intercellular adhesion, lymphocyte signaling, cytotoxicity and lymphokine secretion mediated by cytotoxic T-lymphocyte (CTL) and NK cells. CD226 is a cell surface receptor for NECTIN2 and upon ligand binding, stimulates T-cell proliferation and cytokine production, including that of IL2, IL5, IL10, IL13, and IFNG. Human CD226 shares 53%, 51%, 61% and, 94% sequence homology with mouse, rat, cow, and monkey, respectively.

Biological function summary
CD226 is involved in immune cell activation and receptor-mediated signaling. It aids the recognition and destruction of target cells by NK cells and cytotoxic T lymphocytes through its interaction with ligand-expressing cells. CD226 does not work alone but functions as part of a complex system of co-stimulatory and inhibitory receptors balancing immune responses. Its cooperation with receptors such as CD96 and TIGIT (T cell immunoreceptor with Ig and ITIM domains) modulates the activity of immune effector cells.
Pathways
CD226 takes part in immune signaling pathways that regulate cell-mediated cytotoxicity and inflammation. It is an important component of the immune checkpoint axis interacting closely with proteins like CD96 and TIGIT within these pathways. This interaction helps determine the fate of immune responses influencing the progression or resolution of inflammatory and autoimmune conditions. CD226 plays a role in the NK cell-mediated cytotoxicity pathway and the adaptive immune response.
