Sialic-acid-binding immunoglobulin-like lectin (Siglec) that plays a role in mediating cell-cell interactions and in maintaining immune cells in a resting state (PubMed:10611343, PubMed:11320212, PubMed:15597323). Preferentially recognizes and binds alpha-2,3- and more avidly alpha-2,6-linked sialic acid-bearing glycans (PubMed:7718872). Upon engagement of ligands such as C1q or syalylated glycoproteins, two immunoreceptor tyrosine-based inhibitory motifs (ITIMs) located in CD33 cytoplasmic tail are phosphorylated by Src-like kinases such as LCK (PubMed:10887109, PubMed:28325905). These phosphorylations provide docking sites for the recruitment and activation of protein-tyrosine phosphatases PTPN6/SHP-1 and PTPN11/SHP-2 (PubMed:10206955, PubMed:10556798, PubMed:10887109). In turn, these phosphatases regulate downstream pathways through dephosphorylation of signaling molecules (PubMed:10206955, PubMed:10887109). One of the repressive effect of CD33 on monocyte activation requires phosphoinositide 3-kinase/PI3K (PubMed:15597323).

Human CD33 ELISA kit (ab283542) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Human CD33 protein in human serum, plasma and cell culture extract samples. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human CD33 with 11.39 pg/mL sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Human CD33 is a putative adhesion molecule of myelomonocytic-derived cells that mediates sialic-acid dependent binding to cells. Human CD33 preferentially binds to alpha-2,6-linked sialic acid. The sialic acid recognition site may be masked by cis interactions with sialic acids on the same cell surface. In the immune response, it may act as an inhibitory receptor upon ligand induced tyrosine phosphorylation by recruiting cytoplasmic phosphatase(s) via their SH2 domain(s) that block signal transduction through dephosphorylation of signaling molecules. Human CD33 Induces apoptosis in acute myeloid leukemia (in vitro).

The CD33 standard curve was prepared as described in Section 10. Raw data values are shown in the table. Background-subtracted data values (mean +/- SD) are graphed.
Interpolated concentrations of native CD33 in human serum and plasma.
The concentrations of CD33 were measured in duplicates, interpolated from the CD33 standard curves and corrected for sample dilution. Undiluted samples are as follows: serum 100%, plasma (citrate) 50%, plasma (EDTA) 50% and plasma (heparin) 100%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean CD33 concentration was determined to be 1659.34 pg/mL in serum, 1253.61 pg/mL in plasma (citrate) and 1395.24 pg/mL in plasma (EDTA) and 1288.95 in plasma (heparin).
Interpolated concentrations of native CD33 in human U937 extract, THP-1 extract, and HL-60 extract based on a 500 μg/mL extract load.
The concentrations of CD33 were measured in duplicate and interpolated from the CD33 standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean CD33 concentration was determined to be 1024.31 pg/mL in U937 extract, 688.05 pg/mL in THP-1 extract and 2363.02 pg/mL in HL-60 extract.
