Serum from eight individual healthy CD63 female donors was measured in duplicate.
Interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean CD63 concentration was determined to be 252 pg/mL with a range of 103 – 408 pg/mL.

Interpolated concentration of native Human CD63 was measured in duplicate at different sample concentrations. Undiluted samples are as follows : Purified MCF-7 exosome standards (ab239691) 25 µg/mL (dilutions made into Sample Diluent NS) and denatured MCF-7 exosome standards (ab239691) 2.5 µg/mL (dilutions made in Sample Diluent NS + Enhancer). The interpolated dilution factor corrected values as a ratio of µg/mL exosome load are plotted (mean +/- SD, n=2).

How the assay works
Human CD63 SimpleStep ELISA®employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Human CD63 antigen, also referred to as Granulophysin, Lysomal-associated membrane protein 3 (LAMP-3), Tetraspanin-30, and Ocular melanoma-associated antigen, is a membrane bound protein encoded by the gene CD63. CD63 belongs to a group of proteins known as the tetraspanin family, which are characterized by the presence of four hydrophobic domains and are found to mediate signal transduction events for the regulation of cell development, activation, growth, and motility. Deficiency of this protein is associated with Hermansky-Pudlak Syndrome, which is an autosomal disorder.

Biological function summary
CD63 participates in various cellular processes including membrane trafficking cell adhesion and signal transduction. It associates with integrins and other tetraspanins forming complexes that influence cellular adhesion and migration. Through its role in exosome formation CD63 contributes significantly to intercellular communication by facilitating the transfer of proteins lipids and RNA between cells.
Pathways
CD63 plays a role in pathways linked to cell proliferation and migration and immune response regulation. Within the integrin-mediated signaling pathway CD63 interacts with proteins such as integrins to promote cell adhesion and migration. CD63 also links to vesicle trafficking pathways collaborating with proteins involved in exocytosis and endocytosis which affect cell membrane restructuring and intracellular transport.
