Serum from ten individual healthy human female donors was measured in duplicate.
Interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean CD66b concentration was determined to be 717.64 pg/mL with a range of 356.71 – 1,292.71 pg/mL.

Interpolated concentrations of native CD66b in human PBMC supernatant samples.
Stimulated samples were treated with 1.5% PHA-M for 46 hours. The concentrations of CD66b were measured in duplicates, interpolated from the CD66b standard curves and corrected for sample dilution. Undiluted samples are as follows : stimulated PBMC supernatant 100% and unstimulated PBMC supernatant 100%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean CD66b concentration was determined to be 263.55 pg/mL in stimulated PBMC supernatant and 151.03 pg/mL in unstimulated PBMC supernatant.

The concentrations of CD66b were measured in duplicates, interpolated from the CD66b standard curves and corrected for sample dilution. Undiluted samples are as follows : serum 50%, plasma (citrate) 50%, plasma (heparin) 50%, and plasma (EDTA) 50%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean CD66b concentration was determined to be 802.67 pg/mL in serum, 833.01 pg/mL in plasma (citrate), 1,208.45 pg/mL in plasma (heparin), and 531.15 pg/mL in plasma (EDTA).

CD66b is a cell surface glycoprotein involved in cell adhesion, cell migration and pathogen binding. It is a member of the CEA superfamily of immunoglobulins. CD66b is exclusively expressed on granulocytes. Soluble CD66b is released in response to bacterial DNA.

Biological function summary
CD66b contributes to immune responses by facilitating leukocyte activation and migration during inflammation. It does not form part of a multi-protein complex but interacts with other cellular components. CD66b promotes neutrophil adhesion and transmigration to sites of infection or injury supporting their role in innate immunity. Additionally it participates in signaling pathways that influence cell survival proliferation and apoptosis in response to environmental cues.
Pathways
CD66b operates within neutrophil-related processes like the immune response and cell adhesion pathways. In the immune response pathway it collaborates with integrins to assist neutrophil extravasation through the endothelium. During inflammatory responses CD66b interaction with integrins and other adhesion molecules influences the intensity and duration of activity at inflammation sites. This interaction highlights its involvement with proteins such as ICAM-1 in mediating firm adhesion necessary for neutrophil migration.
