Interpolated concentrations of spiked human CD74 in serum, plasma (heparin), and plasma (EDTA).
Recombinant CD74 was spiked into the following human samples and diluted in a 2-fold dilution series in Sample Diluent NS. Undiluted samples are 50% serum, plasma (heparin), and plasma (EDTA). The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2).

Recombinant CD74 was spiked into PBMC supernatant stimulated for 46 hours in 1.5% PHA-M and diluted in a 2-fold dilution series in Sample Diluent NS. Undiluted samples are 50% stimulated PBMC supernatant. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2).

Human CD74 ELISA kit (ab284618) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Human CD74 protein in human serum, plasma, and cell culture supernatant samples. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human CD74 with 15.63 pg/mL sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Biological function summary
CD74 facilitates multiple immune regulatory processes. It forms a complex with the MHC II molecules aiding in their proper folding and peptide loading in the endoplasmic reticulum. This complex then travels through the Golgi apparatus towards the lysosomal compartments where antigens are presented to initiate adaptive immune responses. In addition CD74 is involved in signal transduction pathways that regulate cell proliferation and survival influenced by interactions with MIF and other molecules.
Pathways
The role of CD74 extends into both the antigen presentation and MIF signaling pathways. Within the antigen presentation pathway CD74's activity is tightly connected with MHC II and subsequently to CD4+ T cells facilitating the activation of these immune cells. The MIF signaling pathway involves CD74 interacting with CD44 a receptor that further propagates the signaling cascades essential for immune modulation and cell survival.
