Interpolated concentrations of native CD86 in human HDLM-2 cell culture supernatant samples.
The concentrations of CD86 were measured in duplicates, interpolated from the CD86 standard curves and corrected for sample dilution. Undiluted samples are as follows : HDLM-2 supernatant 25%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean CD86 concentration was determined to be 7,134 pg/mL in HDLM-2 supernatant.

Human CD86 ELISA Kit (ab242239) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of CD86 protein in cell culture supernatant. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human CD86 with 3.5 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Biological function summary
CD86 plays a significant role in the immune system by providing secondary signals for T cell activation and differentiation. It is a part of the B7 protein family and forms a complex with CD28 and CTLA-4 on T cells. When CD86 binds to CD28 it sends positive co-stimulatory signals which promote T cell proliferation and cytokine production. On the other hand interaction with CTLA-4 transmits an inhibitory signal which reduces immune response. This dual interaction helps to balance immune activation and tolerance.
Pathways
CD86 takes part in important immune-related signaling pathways particularly the T cell receptor signaling pathway and the PI3K-Akt signaling pathway. Both pathways are fundamental for initiating immune responses. CD86's interaction with CD28 activates downstream signaling cascades including PI3K-Akt which is important for cell survival and growth. Additionally CD86 collaborates with other proteins such as CD80 another co-stimulatory molecule to amplify T cell activation within these pathways.
