Interpolated concentrations of native Complement C6 in Human serum, plasma and saliva samples.

The concentrations of Complement C6 were measured in duplicates, interpolated from the Complement C6 standard curves and corrected for sample dilution. Undiluted samples are as follows : serum 1 : 2000, plasma (citrate) 1 : 2000, plasma (EDTA) 1 : 2000, plasma (heparin) 1 : 2000, and saliva 1 : 2. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Complement C6 concentration, corrected for dilution, was determined to be 21 μg/mL in serum, 19 μg/mL in plasma (citrate), 16 μg/mL in plasma (EDTA), 15 μg/mL in plasma (heparin), and 4 ng/mL (0.004 μg/mL) in saliva.

Serum from ten individual healthy Human male donors was measured in duplicate.

Interpolated, dilution factor-corrected values are plotted (mean +/- SD, n=2). The mean Complement C6 concentration was determined to be 23 μg/mL with a range of 16 – 29 μg/mL.

Human Complement C6 ELISA Kit (ab230934) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Complement C6 protein in cell culture supernatant, cit plasma, edta plasma, hep plasma, saliva, and serum. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human Complement C6 with 71 pg/ml sensitivity.

SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Complement component C6 often called simply C6 is an essential protein in the complement system. It is part of the terminal complement pathway which leads to the formation of the membrane attack complex (MAC). C6 has a molecular weight of about 93 kDa and is mainly produced in the liver. It circulates in plasma and interacts with other complement proteins to form MAC aiding in pathogen elimination and cell lysis.
Biological function summary

The role of C6 is the progression and stabilization of the MAC complex which is part of the innate immune system's defense. C6 collaborates with other complement proteins such as C5b C7 C8 and C9. This cooperation helps penetrate the membrane of pathogenic cells leading to cell destruction. Detecting and quantifying C6 levels is possible using techniques like complement C6 ELISA and C6 graphs valuable for research into immune response mechanisms.

Pathways

The complement component C6 is an important player in the terminal complement pathway and the larger complement activation pathway. Within these pathways C6 interacts closely with proteins like C5b and C9 to facilitate the assembly of the MAC. Component of the membrane attack complex (MAC), a multiprotein complex activated by the complement cascade, which inserts into a target cell membrane and forms a pore, leading to target cell membrane rupture and cell lysis (PubMed : 22267737, PubMed : 22832194, PubMed : 26841837, PubMed : 27052168, PubMed : 30552328). The MAC is initiated by proteolytic cleavage of C5 into complement C5b in response to the classical, alternative, lectin and GZMK complement pathways (PubMed : 30552328, PubMed : 39914456, PubMed : 39814882). The complement pathways consist in a cascade of proteins that leads to phagocytosis and breakdown of pathogens and signaling that strengthens the adaptive immune system (PubMed : 30552328). Together with component C5b, involved in MAC complex assembly : complement C5b and C6 associate with the outer leaflet of target cell membrane, reducing the energy for membrane bending (PubMed : 30552328, PubMed : 32569291).
