Serum from ten individual healthy human male donors was measured in duplicate.

Interpolated dilution factor corrected values are plotted (mean +/- SD n=2). The mean Complement Factor H concentration was determined to be 288 µg/mL with a range of 156.1 – 466.5 µg/mL.

Interpolated concentrations of native Complement Factor H in human saliva and A549 cell culture supernatant samples.

The concentrations of Complement Factor H were measured in duplicates interpolated from the Complement Factor H standard curves and corrected for sample dilution. Undiluted samples are as follows : saliva 2.5% and A549 cell culture supernatant 25%. The interpolated dilution factor corrected values are plotted (mean +/- SD n=2). The mean Complement Factor H concentration was determined to be 179 ng/mL in saliva and 31 ng/mL in A549 cell culture supernatant.

The interpolated dilution factor corrected values are plotted (mean +/- SD n=2). The mean Complement Factor H concentration was determined to be 268 µg/mL in serum 247 µg/mL in plasma (citrate) 231 µg/mL in plasma (EDTA) and 187 µg/mL in plasma (heparin).

How the assay works

Human Complement factor H ELISA Kit, Chemiluminescent (ab327042) SimpleStep Ignite™ ELISA employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step during a 30 min only incubation, significantly reducing assay time. By utilizing a chemiluminscent detection method this improves the sensitivity and dynamic range when compared to traditional colorimetric ELISAs.

Target data

Glycoprotein that plays an essential role in maintaining a well-balanced immune response by modulating complement activation. Acts as a soluble inhibitor of complement, where its binding to self markers such as glycan structures prevents complement activation and amplification on cell surfaces (PubMed : 21285368, PubMed : 21317894, PubMed : 25402769). Accelerates the decay of the complement alternative pathway (AP) C3 convertase C3bBb, thus preventing local formation of more C3b, the central player of the complement amplification loop (PubMed : 19503104, PubMed : 21317894, PubMed : 26700768). As a cofactor of the serine protease factor I, CFH also regulates proteolytic degradation of already-deposited C3b (PubMed : 18252712, PubMed : 23332154, PubMed : 28671664). In addition, mediates several cellular responses through interaction with specific receptors. For example, interacts with CR3/ITGAM receptor and thereby mediates the adhesion of human neutrophils to different pathogens. In turn, these pathogens are phagocytosed and destroyed (PubMed : 20008295, PubMed : 9558116).. (Microbial infection) In the mosquito midgut, binds to the surface of parasite P.falciparum gametocytes and protects the parasite from alternative complement pathway-mediated elimination.
