CRHBP also known as Corticotropin-Releasing Hormone Binding Protein plays a role in regulating the activity of corticotropin-releasing hormone (CRH) by binding to it with high affinity. The binding protein has a molecular mass of approximately 37 kDa. It is mainly found in the brain and pituitary gland. CRHBP controls the actions of CRH by sequestering it preventing CRH from interacting with its receptors. This modulation is essential for maintaining the balance of CRH's activity.
Biological function summary

CRHBP modulates the stress response by binding to CRH and limiting its availability to CRHR1 and CRHR2 receptors. By doing so it reduces the downstream signaling that would be induced by free CRH which affects the release of adrenocorticotropic hormone (ACTH) from the pituitary. This regulation is important for controlling how the body responds to stress. CRHBP does not form a larger protein complex but operates by interacting directly with CRH in the extracellular space.

Pathways

CRHBP functions within the hypothalamic-pituitary-adrenal (HPA) axis which plays an important role in the body's response to stress. By regulating CRH availability CRHBP indirectly influences the cortisol production pathway. This is important because cortisol is an important hormone in modulating metabolism and immune responses. Changes in CRHBP expression or function may alter CRH availability impacting mood regulation and psychological resilience.
