Interpolated concentration of native CYR61 was measured in duplicate at different sample concentrations. Undiluted samples are 100% serum, plasma (citrate), and urine. The interpolated dilution factor corrected values (to neat sample) are plotted (mean +/- SD, n=2). Sample dilutions for serum and plasma are made in Sample Diluent 25BP, and sample dilutions for urine and made in Sample Diluent NS.

Interpolated concentrations of native CYR61 in human HeLa cell extract based on a 250 μg/mL extract load and MDA-MB435S cell extract based on a 500 μg/mL extract load. The concentrations of CYR61 were measured in duplicate and interpolated from the CYR61 standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean CYR61 concentration was determined to be 1,097 pg/mL in HeLa cell extract and 1,717 pg/mL in MDA-MB-435S cell extract.

The concentrations of CYR61 were measured in duplicates, interpolated from the CYR61 standard curves and corrected for sample dilution. Undiluted samples are as follows : milk 100%, saliva 100%, HeLa supernatant 25%, and MDA-MB-453S 25%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean CYR61 concentration was determined to be 1,432 pg/mL in neat milk, 798.2 pg/mL in neat saliva, 8,357 pg/mL neat HeLa cell culture supernatant, and 1,984 pg/mL in neat MDA-MB-453S cell culture supernatant.

How the assay works

Human CYR61 ELISA Kit, Chemiluminescent (ab326749) SimpleStep Ignite™ ELISA employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step during a 30 min only incubation, significantly reducing assay time. By utilizing a chemiluminscent detection method this improves the sensitivity and dynamic range when compared to traditional colorimetric ELISAs.

Target data

Promotes cell proliferation, chemotaxis, angiogenesis and cell adhesion. Appears to play a role in wound healing by up-regulating, in skin fibroblasts, the expression of a number of genes involved in angiogenesis, inflammation and matrix remodeling including VEGA-A, VEGA-C, MMP1, MMP3, TIMP1, uPA, PAI-1 and integrins alpha-3 and alpha-5. CCN1-mediated gene regulation is dependent on heparin-binding. Down-regulates the expression of alpha-1 and alpha-2 subunits of collagen type-1. Promotes cell adhesion and adhesive signaling through integrin alpha-6/beta-1, cell migration through integrin alpha-v/beta-5 and cell proliferation through integrin alpha-v/beta-3.
