Antagonizes canonical Wnt signaling by inhibiting LRP5/6 interaction with Wnt and by forming a ternary complex with the transmembrane protein KREMEN that promotes internalization of LRP5/6. DKKs play an important role in vertebrate development, where they locally inhibit Wnt regulated processes such as antero-posterior axial patterning, limb development, somitogenesis and eye formation. In the adult, Dkks are implicated in bone formation and bone disease, cancer and Alzheimer disease (By similarity).

Human DKK3 ELISA Kit (ab236710) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of DKK3 protein in cit plasma, edta plasma, hep plasma, serum, and cell culture supernatant. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human DKK3 with 34.1 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Sandwich ELISA - Human DKK3 ELISA Kit (ab236710)
Serum from ten individual healthy human female donors was measured in duplicate.
Interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean DKK3 concentration was determined to be 40.4 ng/mL with a range of 27.3 – 65.8 ng/mL.

Interpolated concentrations of native DKK3 in human cell culture supernatant samples. The concentrations of DKK3 were measured in duplicates, interpolated from the DKK3 standard curves and corrected for sample dilution. Undiluted samples are as follows: HUVEC supernatant 12.5%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean DKK3 concentration was determined to be 19.0 ng/mL in HUVEC supernatant.
