Abcam's DKK3 (Dickkopf-3) Human ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Human DKK3 in serum, plasma, and cell culture supernatants.

This assay employs an antibody specific for Human DKK3 coated on a 96-well plate. Standards and samples are pipetted into the wells and DKK3 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Human DKK3 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of DKK3 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Optimisation may be required with urine samples.

DKK3 also known as Dickkopf-related protein 3 or Dickkopf-3 is a member of the Dickkopf protein family. This secreted protein has a molecular mass of approximately 38 kDa. DKK3 is expressed in multiple tissues including heart brain and kidney. It plays a role in modulating signaling pathways by acting as a Wnt signaling pathway antagonist. It's important in the regulation of developmental processes and is often investigated in various cellular contexts.
Biological function summary

DKK3 functions as an inhibitor of the Wnt/beta-catenin signaling pathway. It is not known to be a part of a larger protein complex but likely interacts with specific receptors and co-receptors at the cell surface to modulate signaling events. Antagonizes canonical Wnt signaling by inhibiting LRP5/6 interaction with Wnt and by forming a ternary complex with the transmembrane protein KREMEN that promotes internalization of LRP5/6. DKKs play an important role in vertebrate development, where they locally inhibit Wnt regulated processes such as antero-posterior axial patterning, limb development, somitogenesis and eye formation. In the adult, Dkks are implicated in bone formation and bone disease, cancer and Alzheimer disease (By similarity).
